Background Pyogenic liver abscess (PLA) is a common clinical disease throughout the world with remaining high mortality and morbidity. This study sought to investigate key genes and non-coding RNAs (ncRNAs) associated with the pathogenesis and treatment of sepsis caused by PLA (SLA). Methods We constructed the expression profiles of RNAs by RNA sequencing in the peripheral blood of healthy subjects, SLA and seven day therapy of SLA patients. Then the sequencing data was analysed through integrated bioinformatics strategy. Results 4549 mRNAs, 9808 lncRNAs, 467 circRNAs and 292 miRNAs were differentially expressed (DE) between SLA and control samples. 3199 mRNAs, 6018 lncRNAs, 161 circRNAs and 132 miRNAs were differentially expressed between SLA after 7 days therapy and SLA control subjects. Moreover, DE mRNAs of the former two comparisons were both related to immunity and T cell-mediated immune response; DE lncRNAs were both associated with B cell receptor signaling pathway and Osteoclast differentiation; DE circRNAs were both related to Chagas disease and B cell receptor signaling pathway; DE miRNAs exerts its specific role in the process of SLA by MAPK signaling pathway and Estrogen signaling pathway compared to DE lncRNAs, circRNAs and mRNAs. Finally, lncRNA/circRNA–miRNA–mRNA network was constructed to explore the internal regulating relationships of these ncRNAs and mRNAs. Conclusions Our results may provide a new perspective on differential RNA expression involved in the pathogenesis and treatment of SLA and accelerate the development of novel treatments for this disease.

Sepsis is a severe condition characterized by systemic inflammation. One of the most involved organs in sepsis is the heart. On the other hand, heart failure and dysfunction are some of the most leading causes of death in septic patients. miRNAs are short single-strand non-coding ribonucleic acids involved in the regulation of gene expression on a post-transcriptional phase, which means they are a part of the epigenetic process. Recently, researchers have found that miRNA expression in tissues and blood differs depending on different conditions. Because of this property, their use as serum sepsis biomarkers has also been explored. A narrative review is carried out to gather and summarize what is known about miRNAs’ influence on cardiac dysfunction during sepsis. When reviewing the literature, we found at least 77 miRNAs involved in cardiac inflammation and dysfunction during sepsis. In the future, miRNAs may be used as early sepsis-induced cardiac dysfunction biomarkers or as new drug targets. This could help clinicians to early detect, prevent, and treat cardiac damage. The potential role of miRNAs as new diagnostic tools and therapeutic strategies worth deepening the complex network between non-coding RNA and biological pathways. Additional studies are needed to further investigate their role in sepsis-induced myocardium injury.

We explore miR‐150‐5p in atherosclerosis (AS). The AS model was constructed using Apo E−/− mice with an injection of the miR-150-5p mimic or an inhibitor. Pathological characteristics were assessed using Oil red O staining and Masson staining. Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and Western blot were used to analyze the expressions of microRNA-150-5p (miR-150-5p), STAT1, α-SMA (α-smooth muscle actin) and proliferating cell nuclear antigen (PCNA). Targetscan and dual-luciferase reporter assay were used to analyze the interaction between miR-150-5p and STAT1. The viability, migration, cell cycle and α-SMA and PCNA expressions in oxidized low-density lipoprotein (ox-LDL)-stimulated primary human aortic smooth muscle cells (ASMCs) were assessed using molecular experiments. miR-150-5p was reduced in both AS mice and ox-LDL-stimulated human aortic smooth muscle cells but STAT1 had the opposite effect. The miR‐150‐5p inhibitor alleviated the increase of lipid plaque and reduced collagen accumulation in the aortas during AS. Upregulation of α-SMA and PCNA was reversed by miR-150-5p upregulation. STAT1 was targeted by miR‐150‐5p, and overexpressed miR-150-5p weakened the ox-LDL-induced increase of viability and migration abilities and blocked cell cycle in ASMCs, but overexpressed STAT1 blocked the effect of the miR‐150‐5p mimic. This paper demonstrates that miR-150-5p has potential as a therapeutic target in AS, with plaque stabilization by regulating ASMC proliferation and migration via STAT1.
