Overexpression ofCaWRKY27, a subgroupIIe WRKYtranscription factor ofCapsicum annuum, positively regulates tobacco resistance toRalstonia solanacearuminfection

Abstract: WRKY proteins are encoded by a large gene family and are linked to many biological processes across a range of plant species. The functions and underlying mechanisms of WRKY proteins have been investigated primarily in model plants such as Arabidopsis and rice. The roles of these transcription factors in non-model plants, including pepper and other Solanaceae, are poorly understood. Here, we characterize the expression and function of a subgroup IIe WRKY protein from pepper (Capsicum annuum), denoted as CaWRKY… Show more

“…Since its first identification in sweet potato56, WRKY has been characterized in various plant species and shown to be involved in many biological processes, including biotic and abiotic stress responses789 and plant–pathogen interactions910. In tomato, the effector-triggered immunity against Pseudomonas syringae pv.…”

“…Recently, different types of transcription factors, including WRKY, MYB, and AP2/EREBP, have been induced during the ripening of pepper fruits31. Several CaWRKY s have also been reported to be involved in stress responses in pepper91032. However, there are a very few studies on the potential roles of CaWRKY proteins in fruit maturation.…”

“…WRKY transcription factors have been proven to play important roles in plant defence responses to attacks by several pathogens. CaWRKY27, a WRKY protein from pepper ( Capsicum annum ) positively regulates the stress resistance response to Ralstonia solanacearum infection through modulation of SA‐, JA‐ and ET‐mediated signaling pathways in tobacco ( Nicotiana tabacum ) (Dang et al ). Overexpression of the cotton ( Gossypium hirsutum ) genes GhWRKY39‐1 and GhWRKY40 in tobacco regulates its resistance response to R. solanacearum (Shi et al ).…”

“…By ChIP assay, it was found that, amongst the three typical W‐boxes, only the W3‐box appeared to be bound by CaWRKY40, which was further supported by the data from MST analysis. However, the W3‐box was not found to be bound by CaWRKY27 and CaWRKY58, the two other pepper WRKY TFs implicated in pepper immunity against R. solanacearum by our previous studies (Dang et al ., ; Wang et al ., ); this suggests that the binding of CaWRKY40 to the W3‐box containing the CaC3H14 promoter is selective; however, its possible binding by other WRKY TFs cannot be ruled out at present. We also observed that, on R. solanacearum infection, pepper plants transiently overexpressing CaWRKY40 exhibited significantly enhanced CaC3H14 transcription, whereas TRV:: CaWRKY40 plants exhibited significantly down‐regulated CaC3H14 transcript abundance, compared with control plants.…”
