Overexpression of PCDHB9 promotes peritoneal metastasis and correlates with poor prognosis in patients with gastric cancer

Abstract: Gastric cancer (GC) is one of the most common human cancers. Genes expressed only in cancer tissue, especially on the cell membrane, may be useful biomarkers for cancer diagnosis and therapeutics. In the present study, we focused on the PCDHB9 gene, which encodes the transmembrane protein protocadherin B9. Immunohistochemical analysis revealed that 62 (36%) of 173 GC cases were positive for protocadherin B9. Protocadherin B9 staining was mainly observed on the GC cell membrane. Expression of protocadherin B9 w… Show more

“…Several lines of evidence suggest that some integrins play an essential role in bone metastasis in PCa . Our previous study has shown that protocadherin B9 promotes peritoneal dissemination through ITGA3 , ITGA4 , ITGA5 , and ITGB1 in gastric cancer . These lines of evidence indicate that the interaction between protocadherin B9 and integrins is involved in metastasis.…”

“…We previously demonstrated that PCDHB9 modulated ITGA3, ITGA4, ITGA5, and ITGB1 through NF-κB signaling in gastric cancer. 14 We therefore investigated the effect of PCDHB9 on the expression of ITGA3, ITGA4, ITGA5, ITGB1, and transcription factor p65. The qRT-PCR showed that the overexpression of PCDHB9 induced the expression of ITGA3, ITGA4, ITGA5, and ITGB1 ( Figure S3A).…”

“…Cell-cell interaction is important for cancer cell survival in arduous environments. Protocadherin β-9 may promote the formation of spheroid colonies of gastric cancer cells, and thus aid the survival of cancer cells in the ascites environment and prevent anoikis and apoptosis induced by chemotherapeutic agents (18). Additionally, intraperitoneal mesenchymal cells may support long-distance migration of disseminated tumor cells by maintaining direct contract, as observed with MKN45 gastric cancer cells (19).…”
