Overexpression of Dendritic Cell-Specific Intercellular Adhesion Molecule-3-Grabbing Nonintegrin in Dendritic Cells Protecting against Aspergillosis

Abstract: Background:Dendritic cells (DCs) play an important role in host defense against pathogen infection. DC-specific intercellular adhesion molecule-3-grabbing nonintegrin (SIGN) is a group II C-type lectin receptor and specifically expressed on the surface of DCs. This study aimed to determine whether DC-SIGN affects intracellular signaling activation, Th1/Th2 imbalance and aspergillus immune evasion in aspergillus infection, and explore the application of DC-SIGN-modified DCs in immunotherapy. Methods:DCs were fir… Show more

“…[ 34 , 35 ] For example, after fungal infection, DCs capture the fungal particles and present antigens to T cells, which induce a specific Th1 adaptive immune response to fungus. [ 36 39 ] Pene et al [ 40 ] found that intratracheal administration of DC in mice with compromised immunity infected with Pseudomonas aeruginosa could significantly improve the survival of these mice. The similar results found by Bohannon et al [ 41 ] indicated that DC could accelerate the clearance of Pseudomonas aeruginosa in burned mice.…”

“…The interferon-γ (IFN-γ) pathway is emerging as a key player in host immune response. [ 43 46 ] It plays a dual role in anti-tumor immune response. IFN-γ is produced by tumor-specific T cells, it induces an effective anti-tumor immune response by the following ways: (1) its direct anti-proliferative and pro-apoptotic effects on tumor cells; (2) enhancing the expression of MHC and other molecules to increase tumor antigen presentation; and (3) recruiting other immune cells.…”

“…[ 95 , 96 ] In patients treated with PD-1 antibody, a new homozygous truncation mutation of β2M occurs in drug-resistant cells, resulting in lack of surface expression of HLA class I. [ 46 ] Direct loss of the gene encoding the HLA-C∗08:02 class I molecule was recently described in a colorectal tumor that progressed after initial response to T-cell transfer therapy. These T cells were HLA-C∗08:02-restricted TILs, and thus this mutation directly allowed immune evasion by tumor cells.…”
