Overexpression of AdeABC efflux pump associated with tigecycline resistance in clinical Acinetobacter nosocomialis isolates

Abstract: This study showed that the dissemination of TNAN isolates in Taiwan is attributable mainly to the spread of ST410. The AdeABC efflux pump appeared to play an important role in the tigecycline resistance of A. nosocomialis.

“…In general, these efflux pumps can lead to the development of antibiotic‐resistant strains by decreasing the antibiotic concentration inside the cell [4–7]. AdeIJK , AdeABC , and AdeFGH are the most important efflux pumps known in A. baumanii , which belong to RND family [8]. These RND efflux pumps include a multidrug protein carrier ( AdeB , AdeJ , and AdeG ), which is located in the cytoplasmic membrane, and an additional periplasmic protein ( AdeA , AdeI , and AdeF , also known as a membrane fusion protein), mediating cooperation between the protein carrier and the outer membrane [9–11].…”

“…The expression of AdeFGH and AdeIJK is controlled by AdeL, a LysR-type transcriptional regulator and AdeN, a TetR-like transcriptional regulator, respectively (Xu et al, 2019). Our previous study has demonstrated that the overexpression of the AdeABC efflux pump plays a major role in exporting tigecycline in tigecycline non-susceptible A. nosocomialis (Yang et al, 2019). Type 1 AdeRS TCS was found to be associated with tigecycline non-susceptible in clinical Acinetobacter nosocomialis in isolates (Yang et al, 2019).…”

“…Minimum inhibitory concentrations (MICs) of tigecycline, omadacycline and eravacycline were determined by broth microdilution. The U.S. Food and Drug Administration (FDA) recommendation for tigecycline susceptibility breakpoints of Enterobacteriaceae (susceptible MIC≤2 mg/L; intermediate >2 and <8 mg/L; resistant ≥8 mg/L) was used as the MIC interpretation criteria (Yang et al, 2019). Omadacycline and eravacycline were currently without breakpoints.…”
