“…Under red light, PIF1 is degraded, which stimulates the accumulation of vindoline (Liu et al, 2019). PIF3 upregulates the expression of ADS, CYP71AV1, artemisinic aldehyde Δ11( 13) reductase (DBR2), and aldehyde dehydrogenase 1 (ALDH1), and dramatically increases the content of artemisinin under light in A. annua (Zhang et al, 2019c). AaMYB15 is reported as a negative regulator of artemisinin, it binds to the promoter of AaORA and inhibits the biosynthesis of artemisinin under light in A. annua (Wu et al, 2021).…”

“…So, we prepared a nootkatone biosynthesis module for working in the cytosol, the plant expression vector pcNKT, which harbored VS , VO , and hygromycin resistance gene (Hygr, as a selection marker) driven by CaMV 35S promoter (Figure ). A. annua is very rich in sesquiterpenes; for example, the artemisinin level in leaves is generally 10–20 mg/g dry weight (DW), suggesting that FPP is abundant enough for sesquiterpene biosynthesis in A. annua . Therefore, it is not necessary to fortify FPP formation through overexpressing FPS in the cytosol. The vector pcNKT was used to genetically transform the plants of A. annua for nootkatone production in the cytosol.…”

“…Engineered A. annua plants, including cNKT and pNKT lines, were established and micropropagated according to the methods developed before (Figure A–C). Genomic PCR was used to detect the integration of transgenes into the genome of A. annua plants. The 1636-bp fragment of VO :: NOS , the 1915-bp fragment of VS :: OCS , and the 492-bp fragment specific to hygromycin resistance gene, were simultaneously amplified from positive control (pcNKT).…”

“…Since 2002, Artemisinin‐based combination therapies (ACTs) are recommended to treat malaria by the World Health Organization (WHO) 45 . However, the artemisinin content in A. annua is very low, ranging from 0.01% to 1% of dry weight in leaves, 46 resulting in the disposal of about 99% of the plant. Despite contributing significantly to the field of biomedical and fabric engineering applications, 47 its waste can cause environmental problems if destinated incorrectly.…”
