In the current study, we have synthesized canine recombinant Hsp27 in E. coli and raised hyperimmune serum against the protein in mice. Using the hyperimmune serum, an indirect ELISA was developed to estimate circulating levels of Hsp27 in dogs with various types of mammary neoplasia and to compare their levels with those of tumor-free apparently healthy dogs. The developed assay had a high specificity (83.3%) and sensitivity (96.6%) for a cut-off value of 0.45 with respect to histopathological examination in discriminating healthy dogs from those bearing mammary tumors. Serum Hsp27 levels were found to be significantly elevated in tumor subjects (20.33 ± 1.23 ng/ml) as compared to healthy controls (6.56 ± 0.51 ng/ml) and the increase in the Hsp27 levels was irrespective of mammary tumor histotypes. However, dogs with grade-II tumors had higher Hsp27 levels as compared to grade-I types. Therefore, Hsp27 can be exploited as one of the 'neoplastic signatures' of canine mammary tumors.

Canine mammary gland tumour (cMGT) is the most common tumour in intact female dogs. Surgery is the only effective treatment for cMGT, and dogs with metastasis at the time of diagnosis or those diagnosed at an advanced stage have poorer prognosis. Thus, novel diagnostic biomarkers and therapeutic targets are needed. Neurokinin‐1 receptor (NK‐1 receptor) is involved in cancer progression and has been detected in various malignant tumours including breast cancer in humans. Furthermore, NK‐1 receptor antagonists inhibit cancer progression. We evaluated NK‐1 receptor expression in malignant and benign cMGT compared with that in normal mammary gland tissues and analysed the relationship between the expression of NK‐1 receptor and histopathological type or malignancy grade. Specimens from 34 malignant MGT and 35 benign MGT cases were used for immunohistochemistry and scored according to intensity and percentage. Healthy margins from each tumour were used as internal controls. The scores for NK‐1 receptor intensity, percentage of positive cells and overall immunohistochemistry were higher in malignant MGT than in benign MGT and normal tissue (p < .000). NK‐1 receptor expression was not correlated with either malignancy grade or histopathological type. Expression of the NK‐1 receptor in malignant MGT was higher than that in benign MGT and normal tissues. Thus, NK‐1 receptor could be considered a novel therapeutic target for cMGT. Further studies using other quantitative tests such as western blotting or PCR and the evaluation of substance P in patient tumour tissue or serum are needed.

Background- Mammary tumors are frequently seen in female dogs. Conventionally, these tumors are diagnosed by histopathology. Differentially expressed biomarkers could aid in non-invasive diagnosis of canine mammary tumors but their number is scanty. The present investigation focuses on assessing HSPD1 as a diagnostic biomarker of canine mammary tumors. Methods and results- Expression of HSPD1 transcript was analyzed by SYBR green based real time PCR (qRT-PCR). Gene encoding immuno-dominant region of HSPD1 was cloned using the expression vector pPROEX-HTa and expressed in a prokaryotic system. Upon purification by affinity chromatography, a 60KDa recombinant protein was obtained which was confirmed by Western blotting. Hyperimmune serum was raised against the rHSPD1 in mice and immunohistochemistry was standardized to assess expression of this protein in various histotypes of canine mammary tumors. Elevated level of HSPD1 mRNA expression (4.92 ± 0.65folds) was observed in canine mammary tumors as compared to the healthy mammary tissue. Further, HSPD1 protein expression in 80% of healthy tissues showed mild immunoreactivity whereas, moderate to high expression of HSPD1 was seen in 80% CMT tissue samples. Conclusions- The findings suggest that HSPD1 could act as molecular marker for the diagnosis of mammary tumors in canines and also to assess the response to these subjects to treatment.
