This study compared the efficiency of a five-day or standard (nine-day) progesterone-based regimen combined with equine chorionic gonadotrophin (eCG) in a fixed-time AI (FTAI) protocol for dairy cows. The data examined were derived from 3577 inseminations conducted in three dairy herds. Animals with no estrus signs detected over 21 days were randomly assigned to a PRID-9 or PRID-5 group. Cows in each group received a progesterone intravaginal device (PRID) for 9 or 5 days, respectively, PGF2α and eCG on PRID removal, and GnRH 48 h later. Fixed-time AI was performed 12 h after the GnRH dose. Cows artificially inseminated following spontaneous estrus during the study period were considered as controls. Based on the odds ratio, the likelihoods of animals in PRID-9 in the warm (conception rate [CR] of 22.3%) and cool (32% CR) periods, and control animals in the warm period (26.6% CR) becoming pregnant were reduced (by factors of 0.6, 0.3 and 0.4, respectively) compared with the control animals in the cool period (CR of 43.7%). The risk of a twin pregnancy was higher (51.4%) for cystic PRID-9 cows (by a factor of 3.6) and lower (9.9%) for cyclic PRID-5 animals (by a factor of 0.4) compared with the PRID-9 cyclic cows. These findings indicate that the proposed protocol achieves similar results during the cool or warm season to those obtained when AI is conducted at spontaneous estrus during the cool season. In addition, PRID-5 reduced twin pregnancy compared with PRID-9.

ResumoO objetivo deste estudo foi comparar dois protocolos pré-aspiração folicular associados com PGF2α. Foram utilizadas 14 novilhas da raça Nelore, não gestantes, com escore corporal de 2,5 (escala de 1 a 5). Todos os animais foram submetidos aos dois tratamentos (cross over) para controle da onda de crescimento folicular, um através da ablação de todos os folículos 3 dias antes da aspiração folicular (PAF) e outro por controle hormonal (PCH) com benzoato de estradiol e progesterona injetável. Os dados foram analisados pelo ANOVA e as diferenças significativas pelo teste de Tukey, com nível de significância de 5 %. O protocolo controle hormonal (PCH) mostrou-se vantajoso por propiciar maior média de folículos e oócitos, além de redução no número de corpos lúteos evitando também o procedimento extra de aspiração prévia para ablação dos folículos. Conclui-se que o protocolo hormonal (PCH) apresentado mostra-se uma alternativa viável para tratamentos pré-aspiração, havendo benefícios adicionais pela ausência de corpo lúteo no momento da aspiração folicular. Nelore heifers (n=14), not pregnant, body score of 2.5 (scale of 1 to 5) were used. All animals were submitted to two treatments (cross over) to control the follicular wave of growth, one by follicular ablation 3 days before the aspiration (PAF) and another by hormonal control with estrogen and injectable progesterone (PCH). Data were analyzed by ANOVA and significant differences were compared by Tukey test, with significance level of 5%. The hormonal control protocol (PCH) provided advantages as higher average of follicles and oocytes, reduction in the number of corpus luteum and one procedure less -the follicle ablation. We concluded that the hormone control protocol (PCH) is a viable alternative for preaspiration treatments, with additional benefits for the absence of corpus luteum at the time of follicular aspiration.

ABSTRACT:The objective of this study was to develop a superovulatory program based on the synchronization of follicular waves with GnRH which could be applied regardless of the stage of the oestrous cycle. In this experiment, GnRH was given to 30 heifers in lactation between Days 0 and 7 (n = 13), 8 and 12 (n = 12), 13 and 16 (n = 5) of the oestrous cycle. Twenty-four heifers were used as controls and did not receive any GnRH. All follicles ≥ 6 mm were punctured 4 days after GnRH treatment in treated animals and between Days 8 and 12 of the oestrous cycle in control heifers. Two days after the follicular puncture, all heifers were superstimulated with 160 mg Folltropin-V given twice daily over 2 days. Oocytes were collected 42 h after the last FSH treatment. The oocytes were subjected to IVM/IVF and the developmental competence of embryos was compared. In vitro production of embryos was affected only by the stages of the oestrous cycle when the GnRH treatment was given and not by the GnRH treatment. No difference (P > 0.1) in the mean number of oocytes, cleavage and embryo production was noted between the control animals and the animals treated with GnRH in the late phase of the oestrous cycle. The mean number of blastocysts was higher (P < 0.05) in heifers treated with GnRH in the mid and the late phase of the oestrous cycle than in the early phase. In conclusion, the in vitro production of embryos was compromised in the present study with heifers following the follicular synchronization with GnRH. This procedure is advantageous for the in vitro production of bovine embryos since the spontaneous oestrus is eliminated. However, more investigations are needed to increase the competence of oocytes obtained following this procedure.
