The ovarian follicle that contains one single oocyte is the fundamental functional tissue unit of mammalian ovary. Therefore, isolation and in vitro culture of ovarian follicles to obtain fertilizable oocytes are regarded as a promising strategy for women to combat infertility. In this communication, we performed a brief survey of studies on microfluidic encapsulation of ovarian follicles in core-shell hydrogel microcapsules for biomimetic 3D culture. These studies highlighted that recapitulation of the mechanical heterogeneity of the extracellular matrix in ovary is crucial for in vitro culture to develop early pre-antral follicles to the antral stage, and for the release of cumulus-oocyte complex (COC) from antral follicles in vitro. The hydrogel encapsulation-based culture system and the microfluidic technology may be invaluable to facilitate follicle culture as a viable option for restoring women’s fertility in the clinic.

The aims of this study were to verify the effects of Epidermal Growth Factor (EGF) on the morphology, primordial follicle activation, growth and proliferation of granulosa cells of ovine follicles cultured in situ, as well as the effect of a PI3K inhibitor on the follicular activation. Ten ovine ovaries were divided into fragments, being one fixed for histological analysis (fresh control). The remaining fragments were cultured for 7 days in control medium (α-MEM + ) alone or supplemented with EGF (1, 10, 50, 100 or 200 ng/mL). Follicles were classified as normal or atretic, as primordial or growing, and the oocyte and follicle diameters were measured. PCNA immunohistochemistry was performed in the fresh control and in treatment that showed the best results for follicular activation. Pharmacologic inhibition of PI3K activity was performed through pretreatment in media added with 50 µM LY294002 for 1 h. The percentage of normal follicles decreased (P < 0.05) after 7 days of culture in all treatments compared to the fresh control. A significant reduction in the percentage of primordial follicles and an increase (P < 0.05) in the growing ones were observed in all treatments compared to fresh control. Furthermore, both the control medium and 1 ng/mL EGF promoted an increase (P < 0.05) in follicular activation compared to other EGF treatments. The PCNA-positive cells in the EGF treatment were higher (P < 0.05) than in fresh control and α-MEM + . Pretreatment of ovarian tissue with PI3K inhibitor significantly inhibited (P < 0.05) α-MEM + -stimulated primordial follicle activation, but had no effect on EGF-stimulated activation (P > 0.05). In conclusion, PI3K pathway mediates the in vitro spontaneous activation of sheep primordial follicles. Moreover, EGF may act indirectly on follicular activation by promoting granulosa cell proliferation at 1 ng/mL, and EGF inhibited follicle activation in concentrations similar or higher than 10 ng/mL.

The objective of this study was to determine the effects of TNF-α and IL-1β on development and survival of bovine secondary follicle culture in vitro for 18 days. Secondary follicles (~0.2 mm) were isolated from ovarian cortex and individually cultured at 38.5°C, with 5% CO in air, for 18 days, in TCM-199 alone (cultured control) or supplemented with 10 ng/ml IL-1β, 10 ng/ml TNF-α or both TNF-α and IL-1β. The effects of these treatments on growth, follicular survival, antrum formation, viability, ultrastructure and mRNA levels for GDF-9, c-MOS, H1foo and Cyclin B1 were evaluated. The results showed that addition of TNF-α to culture medium increased follicular diameter and rate of antrum formation, whereas that of IL-1β and a mixture of IL-1β and TNF-α did not do so. Ultrastructural analysis showed that, among the tested cytokine treatments, follicles cultured in the presence of TNF-α had the best-preserved oocytes and granulosa cells. The presence of TNF-α, IL-1β or both did not influence the expression of mRNAs analysed. In conclusion, in contrast to IL-1β, TNF-α promotes growth of and antrum formation in in vitro cultured bovine secondary follicles, while their ultrastructure and viability were maintained.
