Ovarian cancer stem cell-specific gene expression profiling and targeted drug prescreening

Abstract: Cancer stem cells, with unlimited self-renewal potential and other stem cell characteristics, occur in several types of cancer, including ovarian cancer (OvC). Although CSCs can cause tumor initiation, malignant proliferation, relapse and multi-drug resistance, ways to eliminate them remain unknown. In the present study, we compared ovarian cancer stem cell (OVCSC) expression profiles in normal ovarian surface epithelium and ovarian cells from patients with advanced disease to identify key pathways and specifi… Show more

“…reported ovarian cancer stem cells (CSCs) for the first time [26]. Ovarian CSCs behave like “seeds” and play a crucial role in tumor initiation, growth, invasion, metastasis, and recurrence [27]. Although the concept of cancer stem cells is still controversial, it is widely accepted that clonal evolution exists in cancer initiation and progression [28].…”

“…Increasing evidence has highlighted the existence of CSCs and their role in tumor relapse and drug resistance (14,15). The first successful isolation of CSCs from a solid tumor occurred in 2003 (16), demonstrating the existence of CSCs in cancers.…”

“…In a genomic study examining a panel of ovarian cancer cells ALDH expression as a feature of stemness was again confirmed suggesting disulfiram as a stemness defeating drug for that type of cancer [155]. As for other cancers, the subset of high ALDH expressing cells was associated with stem cell attributes and disulfiram again, as predicted, defeated much of the stem cell functions, both in vitro and in a xenograft model [156].…”
