Outlining the Grb2 interactome data and its interacting partners in HEK293 cells in absence and presence of epidermal growth factor

Abstract: Growth factor receptor-bound protein 2 (Grb2) is an adaptor protein involved in the signal transduction pathways. This dataset enlists proteins which interact with Grb2 in the presence and absence of a mitogenic stimulus. Grb2 expressing HEK293 cells were cultured in light and heavy labeled SILAC media. Normal lysine and arginine were incorporated as light labels while 8 and 10 Da heavier labels of respective isotopes were used for heavy labeling. While light labeled cells were used to enrich basal Grb2 intera… Show more

“…FOXM1 stimulates indoleamine 2,3 dioxygenase (IDO2) which stimulates T-reg cells which are immunosuppressive and induce tumour tolerance. CB1R also interacts directly with the pro-oncogenic Growth Factor Receptor Bound Protein 2 (GRB2) [ 100 102 ] and stimulates its interactome which signals activation to many oncogenic nuclear genes including RAS [ 99 ]. CB1R and IDO2 also stimulate angiogenesis and the ingrowth of new vessels to the developing tumour [ 99 ].…”

“…Here we report a key addition to this group through demonstrating the direct interaction between GRB2 and AGO2, and the involvement of this complex in the regulation of miRNA expression. Mapping of the GRB2 interactome has previously revealed association with both miRNA biogenesis factors and mediators of gene silencing, suggesting GRB2 may play a larger role in miRNA-mediated translational repression than has previously been identified 51 . The herein described GRB2 interaction is the first to be reported for the AGO2 PAZ domain.…”

“…GRB2 is an adaptor protein ( 24 , 25 ). It contains a conformationally conserved SH2 domain (hereafter referred to as SH2 GRB2 ) flanked by N-terminal and C-terminal SH3 domains (nSH3 and cSH3) ( Fig.…”
