Osteopontin (OPN)/SPP1: from its biochemistry to biological functions in the innate immune system and the central nervous system (CNS)

Abstract: Osteopontin (OPN) is a multifunctional protein, initially identified in osteosarcoma cells with its role of mediating osteoblast adhesion. Later studies revealed that OPN is associated with many inflammatory conditions caused by infections, allergic responses, autoimmunity, and tissue damage. Many cell types in the peripheral immune system express OPN with various functions, which could be beneficial or detrimental. Also, more recent studies demonstrated that OPN is highly expressed in the central nervous syst… Show more

“…Spp1 is a multifunctional protein involved in various biological processes, including cell adhesion, migration, immune regulation, and the in ammation cascade [12] . In the eld of cardiovascular research, the role of Spp1 is increasingly recognized due to its association with the onset and progression of various cardiovascular diseases.…”

“…At P5, but not P6, CCL2 transcript levels were highly increased by ethanol. SPP1, which is also termed osteopontin, is a secretory molecule expressed by a variety of immune cells which has functions including immunomodulation, chemotaxis, and cell adhesion ( Lin et al, 2022 ). SPP1 plays a role in alcohol liver disease ( Apte et al, 2005 ; Seth et al, 2006 ; Lebel et al, 2008 ; Das et al, 2022 ).…”

“…The cDNA was a murine homologue of rat bone OPN identified three years earlier [3]. It has been known for a long time that OPN is involved in immune processes and expressed by many different types of immune cells [111]. The concentration of OPN in milk is orders of magnitude higher than anywhere else in the body, which could indicate that OPN plays a role either as part of the innate immune system in milk, or as a modulator of immune responses in neonates and infants.…”
