Osteonecrosis of the Jaw and Oral Health–Related Quality of Life After Adjuvant Zoledronic Acid: An Adjuvant Zoledronic Acid to Reduce Recurrence Trial Subprotocol (BIG01/04)
Abstract:Adjuvant zoledronate used in the intensive schedule studied in the AZURE trial is associated with a low incidence of ONJ but does not seem to adversely affect Oral-QoL.
“…1,2,3 One of the predisposing factors for MR-ONJ is a history of surgical trauma to the jaw bone. 4 Denuded necrotic bone after implant surgery or extraction of a tooth has frequently been associated with the clinical presentation of MR-ONJ.…”

“…In the real-world setting, the rate of adverse effect of bone-targeted therapies appears higher than that reported in clinical trials [38,39]. dose dependent; an 8-mg dose of zoledronic acid caused more renal toxicity than the 4-mg dose in phase III clinical trials of both breast and prostate cancer [21,22].…”

“…All patients entered the model in the "On therapy" state after surgical resection and could transition to other states based on event rates derived from the literature and described in the Event Rates subsection (next). Only patients treated with za could transition into the "Osteonecrosis of the jaw" state 18 . All patients were subject to state-specific and age-specific background mortality based on Canada life tables 19 .…”

“…Osteonecrosis of the jaw necessitating discontinuation of za developed in 1.6% of the patients in azure ( Table i) 18 . Of those patients, 46% required minor surgical procedures 25 .…”
