Osteogenic potential of dualblocks cultured with human periodontal ligament stem cells: in vitro and synchrotron microtomography study

Abstract: The chosen method, supported by phase contrast micro-CT analysis, successfully and quantitatively monitored the early stages of bone formation and the rate of the bioscaffold resorption in basal and differentiating culture media.

“…This finding mismatches with previous results obtained on not xeno-free media [16], where the mineralized clusters were detectable just starting from the second week of culture in both the culture media, and were mainly concentrated in the trabecular portion.…”

“…In fact, the phase-contrast X-ray imaging provides a substantially enhanced contrast resolution for both soft and mineralized tissues compared to conventional X-ray tube microCT experimental setups. The level of detail within the tomographic images is sufficiently consistent to allow the discrimination between the different levels of mineralization in engineered bone sites [16]. Indeed, the edges and the interfaces between collagenated and mineralized bone (or between areas at different mineralization degrees) could be clearly discernible because of the beam refraction occurring at the edges between tissues with different refraction indexes.…”

“…Finally, the formation of mineralized nodules was observed under a stereomicroscope (SZXl6, Olympus, Japan). The quantification of mineralization was performed as described previously [26]. Briefly, 10% cetylpyridinium chloride was added to each well to dissolve the deposition, and the solution was then collected.…”

“…For Alizarin red quantification, 1 ml of 10% cetylpyridinium chloride was added to each well as described previously. 37 Light absorbance of the extracted dye was determined at 562 nm using a spectrophotometer (NanoDrop Technologies, DE, USA). The experiments were performed 3 times.…”
