Osteogenic Potential of Biosilica on Human Osteoblast-Like (SaOS-2) Cells

Abstract: Biosilica is a natural polymer, synthesized by the poriferan enzyme silicatein from monomeric silicate substrates. Biosilica stimulates mineralizing activity and gene expression of SaOS-2 cells. To study its effect on the formation of hydroxyapatite (HA), SaOS-2 cells were grown on different silicatein/biosilica-modified substrates (bone slices, Ca-P-coated coverslips, glass coverslips). Growth on these substrates induced the formation of HA nodules, organized in longitudinal arrays or spherical spots. Nodules… Show more

“…The technical details for the qRT-PCR have been given earlier (Wiens et al, 2010). The following two genes were selected to determine the activation state of the human chondrocytes; SOX9 SRYbox 9 (NM_000346) with the primer pair Fwd: 5′-TGCTGCTGGGAAACATTTGCAC-3′ (nt 2762 to nt 2783 ) and Rev: 5′-GGGCACTTATTGGCTGCTGAAAC-3′ (nt 2901 to nt 2879 ; 140 bp) and second COL3A1 (collagen type III alpha 1; NM_000090) with the primer pair Fwd: 5′-ATTCCTTCGACTTCTCTCCAGCC-3′ (nt 4182 to nt 4204 ) and Rev: 5′-GTGTTTCGTGCAACCATCCTCC-3′ (nt 4377 to nt 4356 ; 196 bp).…”

“…The cultures were started with 3×10 4 cells/well in a total volume of 3 ml. After an initial incubation period of 3 days, the cultures were continued to be incubated for a total of 5 days in the absence or presence of the MAC, comprising 5 mM β-glycerophosphate, 50 mM ascorbic acid and 10 nM dexamethasone to induce biomineralization (Wiens et al, 2010b). The polyP sample, aCa-polyP-NP, was added to the cultures at concentrations between 0 and 30 µg/mL, as indicated.…”

“…In addition, the existence of endopolyphosphatases has been analytically proven, but they have not yet been purified (Müller et al, 2015a). The released orthophosphate can act, similar to inorganic phosphate, as a source for the mineralization process of cells in culture (Wiens et al, 2010b;Müller et al, 2011); likewise, the orthophosphate can inhibit carbonic anhydrase, the seed-forming enzyme during bone mineral deposition in vitro (Müller et al, 2013). Inorganic phosphate can replace β-glycerophosphate in the mineralization-activation cocktail (MAC, usually comprising β-glycerophosphate, ascorbic acid and dexamethasone) in vitro (Müller et al, 2011).…”

“…6 Orthosilicic acid (Si[OH] 4 ), a soluble form of silicon, does not affect the synthesis of type I collagen for bone tissue regeneration, whereas an Silicon nitride is resistant to bacterial infection during bone regeneration, 9 and silicon increases the proliferation and osteogenic differentiation of osteoblast-like cells. 10 In addition, silica-based bioglass ceramics and biosilicates enhance bone-like structure formation.…”
