Osteoarthritis Year in Review 2016: biomarkers (biochemical markers)

Abstract: There has been steady progress in osteoarthritis (OA) biomarker research in 2016. Several novel biomarkers were identified and new technologies have been developed for measuring existing biomarkers. However, there has been no "quantum leap" this past year and identification of novel early OA biomarkers remains challenging. During the past year, OARSI published a set of recommendations for the use of soluble biomarkers in clinical trials, which is a major step forward in the clinical use of OA biomarkers and bo… Show more

“…Protein targets proposed to generate the antibody bead arrays were selected based on thorough mining of experimental evidence in the literature of rheumatic diseases 3638 and in previous in-house efforts in the field of osteoarthritis using mass spectrometry analyses 24, 28, 39 . The antibody set was finally designed according to the antibody availability within the Human Protein Atlas (HPA) 40 .…”

“…These include predictive markers, target occupancy markers and pharmacodynamic or target engagement markers. Although predictive biomarkers may be of greatest use in a clinical trial, prognostic biomarkers will also play a part, and much of the work done to date by others has been in the prognostic biomarker category 149 . For example, in regard to osteoarthritis, many inflammatory biomarkers have been associated with disease progression.…”

“…These factors show remarkable similarity to those found in AC, including MMP 1‐3 and 7‐14, although their expression levels may differ from those in the NP (see Table ). Additionally, the ECM in the NP and AC is also cleaved by ADAMTSs 4 and 5 …”
