Osmolyte regulation by TonEBP/NFAT5 during anoxia-recovery and dehydration–rehydration stresses in the freeze-tolerant wood frog (Rana sylvatica)

Abstract: BackgroundThe wood frog, Rana sylvatica, tolerates freezing as a means of winter survival. Freezing is considered to be an ischemic/anoxic event in which oxygen delivery is significantly impaired. In addition, cellular dehydration occurs during freezing because water is lost to extracellular compartments in order to promote freezing. In order to prevent severe cell shrinkage and cell death, it is important for the wood frog to have adaptive mechanisms for osmoregulation. One important mechanism of cellular osm… Show more

“…Enhanced chemiluminescent and Coomassie blue‐stained immunoblots were visualized using the Chemi‐Genius BioImaging system (Syngene, Frederick, MD), and densitometric analysis was performed using the associated Gene Tools software. Immunoblot chemiluminescent band intensities were standardized against a group of Coomassie blue‐stained bands in the same lane that showed constant expression levels to correct for any minor variations in sample loading as previously published . The fold changes for the 24‐hour anoxia exposure and 4‐hour aerobic recovery samples are reported relative to the control group by setting the control group to 1.…”

“…There are five members in the NFAT family, where NFATc1‐c4 are regulated by calcium influx and NFATc5 is regulated by osmotic stress . Our laboratory has previously studied the role of NFAT5 over the freeze/thaw, anoxia/reoxygenation, and dehydration/rehydration cycles in the wood frog, therefore prompting the current study to investigate the role of NFATc1‐c4 over the anoxia/reoxygenation cycle. Given that NFATc1‐c4 are regulated by calcium levels in the cell, it is important to understand how calcium levels are influenced by anoxia.…”
