Orthobunyavirus Entry into Neurons and Other Mammalian Cells Occurs via Clathrin-Mediated Endocytosis and Requires Trafficking into Early Endosomes

Abstract: c La Crosse virus (LACV) is a leading cause of pediatric encephalitis and aseptic meningitis in the midwestern and southern United States, where it is considered an emerging human pathogen. No specific therapies or vaccines are available for LACV or any other orthobunyaviruses. Inhibition of LACV entry into cells is a potential target for therapeutic intervention, but this approach is limited by our current knowledge of the entry process. Here, we determined that clathrin-mediated endocytosis is the primary me… Show more

“…These data suggest that LACV is not harbored by endothelial cells. Finally, LACV is internalized via a clathrin-mediated mechanism [18] while transcytosis through BCECs typically utilizes a caveolin-1 mediated mechanism [36]. Thus, LACV likely does not cross the BBB by mechanisms involving transcytosis or direct endothelial cell infection.…”

“…Most bunyaviruses studied to date utilize clathrin-mediated endocytosis to gain access to the cell interior (22,25,38,39,55,58). For UUKV, a clathrin-independent pathway has been suggested as an alternative route (39).…”

“…Heparan sulfate has been implicated in RVFV attachment (4). Sensitivity to clathrin and dynamin-2 perturbation implies a role for clathrin-mediated endocytosis in uptake and internalization (5)(6)(7)(8)(9).…”
