Ornithine/phosphate antiport in rat kidney mitochondria

Abstract: ['4C]Ornithine uptake by rat kidney mitochondria has been investigated according to the stop inhibitor method by using praseodimium chloride as an inhibitor. The existence of an ornithine/Pi exchange was found occurring with 1 : 1 stoichiometry. Both uptake and efflux follow first-order kinetics with a k of 2.4 min-'. Uptake increases with increasing pH. The activation energy for the process is 58.6 kJ/mol and Qlo is 2.6. Ornithine/P, exchange is electrical and energy-dependent, as suggested by the sensitivity… Show more

“…For example, differential expression of the arginase isoenzymes could provide a means to preferentially direct ornithine either to proline or glutamate synthesis via OAT or to polyamine synthesis via ODC (Figure 2). If so, this would imply that ornithine does not rapidly equilibrate between cytosolic and mitochondrial compartments, despite the existence of transporters via which ornithine can traverse the mitochondrial membrane [204][205][206][207][208]. So far as we are aware, no experiments to determine the effect of arginase localization on the metabolic fates of ornithine have been performed.…”

“…Maleimides (N-ethylmaleimide and eosin 5-maleimide) and 5,5'-dithiobis(2-nitrobenzoate) also strongly inhibited the carrier activity, although less efficiently than the mercurials. PrC13, found to inhibit an ornithine/phosphate exchange in kidney mitochondria [9], inhibited the purified ornithine/ citrulline carrier but only at high concentrations. The data of Table 5 also show that the reconstituted ['4C]ornithine exchange was inhibited by externally added citrulline and lysine.…”
