Ornithine Decarboxylase Inhibition: A Strategy to Combat Various Diseases

Abstract: Ornithine decarboxylase is the first enzyme in the polyamine biosynthetic pathway. It is the rate-limiting enzyme which is included during the change of ornithine to putrescine which is the first polyamine. Polyamines (putrescine, spermidine, spermine) are natural and synthetic compounds which contain two or more amino group. Polyamines are highly implicated in cellular functions such as cell growth & multiplication, DNA stabilization, gene transcription and translation, ion-channel activity, etc. Elevated lev… Show more

“…For instance, in addition to NO synthesis it may affect the urea cycle as well as protein and polyamine synthesis, potentially affecting ECM treatment outcome. Polyamines, for instance, have been shown to be important for malaria parasites and also in the physiology and metabolism of the nervous system 46,47 . Both human and mouse cerebral malaria cause depletion of arginine, citrulline and ornithine, due to a marked decrease in arginine appearance 9 .…”

“…While quiescent cells show low ODC activity and putrescine levels, actively dividing cells exhibit increased ODC activity [ 8 , 9 , 10 , 11 ]. Enzyme activity is high during late G1 phase before DNA synthesis begins and again in the G2/M transition phase [ 8 , 9 , 10 , 11 ]. An essential downstream function of the polyamine pathway is the hypusination and activation of eukaryotic initiation factor eIF5A, vital for protein synthesis and cellular proliferation [ 12 ].…”

“…The polyamine biosynthetic pathway is initiated by enzyme ornithine decarboxylase (pyridoxal-5'-phosphate-dependent enzyme). Therefore, inhibition of ornithine decarboxylase may be the essential approach for the management of cancer and other various diseases: malaria or African sleeping sickness, neglected tropical diseases [43][44][45][46][47]. This situation prompts an intensive search for new ornithine-based pharmaceuticals.…”
