ORIGINAL ARTICLE: Pathomorphologic and Immunohistochemical Study on the Devastation of Rat Embryos by Antiphospholipid Antibody Positive Serum

Abstract: These findings support the existence of a novel factor in the APLA-positive serum, causing a detrimental effect to the conceptus, without any relation to the antiphospholipid antibodies.

“…36 Interestingly, a detrimental effect on rat embryo culture of locally injected under 30 kDa fraction from boiled aPL positive serum, that was abolished when purified IgG was injected, may indicate another as-yet unidentified factor leading to fetal loss. 37 Thus, the use of whole sera may suggest a more physiologic condition where multiple factors interplay together.…”

“…Halperin et al offered a novel factor in APS boiled and denaturated sera that is lower than 30 kDa and has a detrimental effect directed against embryonic cells without damage to the trophoblast cells. 37 In addition, during pregnancy of SLE/APS women the embryo develops from the beginning in a detrimental environment, here the embryos developed in a normal setting until 10.5 days, and were under stressful condition only for 28 h. A longer culture period may be needed in order to create a more stressful atmosphere as to get a more pronounced damaging effect.…”

“…They concluded that one possible mechanism of stillbirth as a result of anti-cardiolipin antibodies (aCL) was the induction of thrombotic events in the placenta. In addition to thrombosis, Halperin et al [30]. reported that the injection of human aPL positive sera into the uterus of pregnant rats rapidly and apoptotically destroyed intrauterine embryos, suggesting the existence of a novel factor in the aPL positive serum unrelated to aPL that exerts a detrimental effect to the conceptus without any relation to the aPL.…”
