Origin of 5‐hydroxytryptamine‐induced hyperpolarization of the rat superior cervical ganglion and vagus nerve

Abstract: 1 5-Hydroxytryptamine (5-HT)-induced membrane potential changes were recorded extracellularly from rat superior cervical ganglia (SCG) and cervical vagus nerves in vitro. 2 On the SCG, low concentrations of 5-HT (I x 10-8-3 x 10-7M) induced concentration-related hyperpolarization responses. Higher concentrations of 5-HT (I x 10-6_1 X 10-4M) induced complex responses which typically consisted of an initial hyperpolarization, followed by a depolarization and subsequent after-hyperpolarization. The depolarization… Show more

“…In the guinea-pig vagus nerve the afterhyperpolarizations, like 5-HT induced depolarizations were blocked by 5-HT3 antagonists. This suggests that as in the rat vagus nerve, after-hyperpolarization is a consequence of depolarization and is not mediated by a different 5-HT receptor type (see Ireland, 1987).…”

“…5-HT3 receptor antagonists caused rightward shifts of the concentration-depolarization response curves to 5-HT (see Table 2, Ireland &Butler et al, 1988). For some compounds (MDL 72222, ICS 205-930, GR65630, granisetron, zacopride and GR67330) the rightward shifts were accompanied by a fall in maximum response.…”

“…The method for mounting the preparations in the baths and the techniques for recording and drug application are described in the companion paper (Ireland, 1987). As in this previous study, the temperature of each preparation was maintained at 27 ± 1VC.…”

“…This hyperpolarization is not a consequence of 5-HT-induced depolarization and, unlike the depolarization, does not result from the activation of 5-HT3 receptors since it is resistant to blockade by MDL 72222 and metoclopramide (Ireland, 1987;. However, a more precise characterization of the receptor mediating the hyperpolarization response has not been published.…”
