Origin and functional specializations of DC subsets in the lung

Abstract: Lung DC bridge innate and adaptive immunity, and depending on the context, induce Th1, Th2 or Th17 response, to optimally clear infections. Conversely, lung DC can also prevent overt and harmful immune responses to harmless inhaled antigens via induction of Treg cells or via induction of neutralizing mucosal IgA antibodies. Here, we propose that these functions are not the result of a single population of DC, and instead, subsets of DC perform specialized functions.

“…Similarly, the involvement of intraepithelial conventional CD11b -DCs (cDCs), lamina propia CD11B high cDCs and plasmacytoid (pDCs) without danger signals add to this anti-inflammatory state of the immune system [16]. As the Concentrations of the polymorphonuclear leucocytes (PMN) mobilizer granulocyte colony-stimulating factor (G-CSF) were determined by enzyme-linked immunosorbent assay (ELISA).…”

“…In contrast, professional immune cells, especially alveolar macrophages and supported by type II epithelial cells, are located in the alveoli and with their PRRs they can rapidly recognize the PAMPs of pathogens being inhaled or aspirated to the periphery of the lungs [3,4,16,17]. The initiated inflammation follows within few hours, primarily with recruitment of PMNs, and influx of humoral factors such as complement, defensins and cytokines, as the alveolar lumen and vascular lumen is within a distance of a few mm.…”

“…Plantinga, Hammad and Lambrecht [67] delve deeply into pulmonary DC to study DC biology at a pivotal mucosal surface. They emphasize that different DC subsets exert different functions, from the induction of Treg specific for environmental antigens to the formation of both protective IgA and allergenic IgE responses.…”
