Orgyia Pseudotsugata Baculovirus p10 and Polyhedron Envelope Protein Genes: Analysis of their Relative Expression Levels and Role in Polyhedron Structure

Abstract: To investigate the regulation of pl0 and polyhedron envelope protein (PEP) gene expression and their role in polyhedron development, Orgyia pseudotsugata multinucleocapsid nuclear polyhedrosis viruses lacking these genes were constructed. Recombinant viruses were produced, in which the pl0 gene, the PEP gene or both genes were disrupted with the fl-glucuronidase (GUS) or fl-galactosidase (lacZ) genes. GUS activity under the control of the PEP protein promoter was observed later in infection and its maximal exp… Show more

“…The mature bigger polyhedron is present in the nucleus or extracellular medium and has an envelope, which confers its defined geometric shape ( Figure 5). The polyhedron envelope or "calyx" seems to have two roles: to prevent the loss of virions, and to avoid polyhedron aggregation (Gross et al, 1994). In the electron micrographs, the polyhedron envelope has a membrane-like structure, as shown in Figure 6.…”

“…Brancalhão et al (2009) classified these polyhedra as immature and mature, respectively; the geometric shape is provided by the envelope. The envelope or "calyx" acts to protect the virions, preventing their loss to the external environment (GROSS et al, 1994) and prevents the aggregation of the polyhedra (LIANG et al, 2013).…”

“…A P10 é uma proteína com massa molecular de 10 kDa, que está envolvida no processo de oclusão das partículas virais e provavelmente na lise celular no final da infecção (Van Oers et al, 1994). Também em estudos com a construção de vírus recombinantes sem o gene p10 foi demonstrado que essa proteína é importante para manter a integridade do poliedro (Vlak et al, 1988;Williams et al, 1989;Gross et al,1994).…”
