Organization of the murine immunoglobulin VH complex in the inbred strains.

Abstract: Deletion mapping analyses have been employed to order the heavy chain variable region (VH) gene families in three inbred murine strains. These nine VH gene families have been positioned with respect to the J558 and 3660 VH families in A/J (Ighe) as follows: 3609‐J558‐(J606,VGAM3‐8,S107)‐3660‐(X24,Q52,7183)‐DH. Maps generated with respect to J558 in the BALB/c (Igha) and C57BL/6 (Ighb) strains are consistent with these results. The organization of the VH complex produced by deletion mapping is quite different f… Show more

“…As previously reported, the VH36-65 probe, VId-130 (32), hybridizes to the VHARS-bearing Eco RI fragment (6.2 kbp) of the Igh`haplotype but does not hybridize to BALB/c (Igh°) genomic DNA. In agreement with the data of Rathbun et al (12), we have demonstrated that the VHARS gene is retained by CXAL 1.40 (Fig . 6), indicating that this gene is 5' of the rearranged VHJ606 gene on the Igh`chromosome .…”

“…Bothwell et al (36) and Givol et al (37) reported similar findings for members of the VJ558 family. Previous analyses of B cell lines for V gene deletions (11,12,38,39) were consistent with a clustered organization of V gene family members. V gene analysis of Igh-recombinant mouse strains constructed by Riblet (40) also indicated a generally clustered organization of VH families (10).…”

“…Family size varies from 1 member (V12) to approximately 70 members (J558) (9,31,43). Although early studies showed that genes encoding members of different families were physically clustered within the Ig heavy-chain locus (9,52), subsequent studies have revealed that there is interspersion of some V H families (45,51) as shown in Fig. 1.…”

“…Differences in the replication times observed between different strains of mice could be due to differences in the relative sizes or order of the VK families or could be due to the degree of interspersion among the different mouse strains. Interspersion has been observed in the human VH (1,39) and VK loci (44) and to a much lesser extent the VH locus in the mouse (2,8,9,37,58,59). In the human kappa locus, a large region of the locus was duplicated (38,54).…”
