Organic Chemicals Adsorbed onto Diesel Exhaust Particles Directly Alter the Differentiation of Fetal Thymocytes Through Arylhydrocarbon Receptor but Not Oxidative Stress Responses

Abstract: Diesel exhaust particles (DEP) were reported to have adverse effects on the immune system of laboratory animals and to induce thymic involution, particularly when exposure occurred during the fetal or lactational period. DEP consist of a carbon core to which many organic compounds are adsorbed, including polyaromatic hydrocarbons (PAHs) and their derivatives (e.g., dioxins and quinones). Although it has been suggested that these organic compounds were responsible for mediating the effects of DEP through their … Show more

“…Analysis of networks found aryl hydrocarbon receptor (AhR)-dependent genes, like the xenobiotic metabolizing enzyme CYP1B1 (GO: 0006805) increased in oral mucosa of smokers 67 . The data confirm a pivotal association of the AhR in the genetical stress response provoked by chemicals occurring also tobacco smoke [68][69][70] .…”

“…Using BMDM derived from AhR −/− mice showed that CARB ULSD- and S100-mediated induction of CYP1A1 and COX-2 depends on a functional expression of the AhR. The role of AhR in mediating the effects of diesel exhaust particles on the differentiation of thymocytes has been emphasized earlier in vitro (Ito et al, 2006; van Voorhis et al, 2013) as well as in vivo (O’Driscoll et al, 2018). The results suggest that S100 and CARB ULSD contain components capable of activating CYP1A1 in an AhR-dependent manner.…”

“…This is due mostly to the longer half-life of H 2 O 2 and its ability to diffuse easier through membranes than other types of ROS [ 129 , 130 ]. Slight variations in ROS content may have deep effects on stem cell fate [ 130 , 131 ]. iPSCs and ESCs exhibit similar ROS levels and mitochondrial mass, but significantly less than in human dermal fibroblasts from which iPSCs were generated.…”
