Orexin and Alzheimer’s Disease: A New Perspective

Abstract: Orexin’s role in human cognition has recently been emphasized and emerging evidences indicate its close relationship with Alzheimer’s disease (AD). This review aimed to demonstrate recent research on the relationship between orexin and AD. Orexin’s role in stress regulation and memory is discussed, with significant findings related to sexual disparities in stress response, with potential clinical implications pertaining to AD pathology. There are controversies regarding the orexin levels in AD patients, but th… Show more

“…Acupuncture Modulates the Orexin System. Neurons in the lateral hypothalamic region produce orexin, which has two subtypes: orexin A (OXA) and orexin B (OXB) [99]. Orexin neurons can project into the hippocampus and the medulla oblongata, and are involved in the regulation of synaptic plasticity and learning and memory.…”

“…One emerging question in this field is whether the deficiency of orexins is protective of AD. It was found that compared with controls, narcolepsy patients showed reduced amyloid deposition and neurodegenerative marker levels [17,18].…”

“…20 Besides, the ApoE4 allele is likely to aggravate sleep disturbance in AD patients. 21 Some neurotransmitters, such as glutamate, 22 serotonin, 23 acetylcholine, 24 and orexin, 25 are changed or imbalanced in AD patients, and these neurotransmitters and their receptors generally are related to sleep and wakefulness. Accordingly, we discussed the evidence about sleep−wake disorders and the mechanism of AD in this review, in which we suggested some possibilities for the treatment and diagnosis of AD.…”
