Orexin-A Levels in Relation to the Risk of Metabolic Syndrome in Patients with Schizophrenia Taking Antipsychotics

Abstract: Our results revealed that the orexin-A level was upregulated in patients with schizophrenia treated with antipsychotics, especially for the group taking less obesogenic antipsychotics. Furthermore, higher orexin-A levels were independently associated with better metabolic profiles. These observations suggest that an upregulation of orexin-A has a protective effect against the development of metabolic abnormalities in patients with schizophrenia receiving antipsychotic treatment.

“…Higher total WBC count was found to be associated in two studies ( 64 , 80 ). Three studies reported male gender to be associated with MetS ( 46 , 74 , 76 ), while four studies found female gender to be a risk factor for MetS ( 54 , 71 , 89 , 91 ). Several studies reported a higher age of patients treated with antipsychotics to be significantly associated with the development of MetS ( 43 , 46 , 55 , 73 , 74 , 85 , 87 , 91 ).…”

“…Literature suggests that nicotine can reduce food intake and body weight ( 104 ) and might therefore reduce the risk of MetS ( 46 , 74 ). Interestingly, in our systematic review, tobacco smoking was found to be positively associated with MetS.…”

“…illustrated that orexin-A is stimulated and upregulated by nonobesegenic antipsychotic drugs. [ 47 ] Also, the high orexin level in patients with schizophrenia treated with antipsychotic drugs is regarded as a protective factor against the development and risk of drug-induced metabolic syndrome. [ 48 ] Furthermore, orexin agonist like modafinil ameliorates cognitive function, attention, and antipsychotic-induced sedation.…”

“…Similarly, orexin antagonists abolish olanzapine and haloperidol effect on midbrain dopaminergic neurons, suggesting that orexin is an important neurotransmitter mediates the action of antipsychotic drugs [46]. As well, Chen et al illustrated that orexin-A is stimulated and upregulated by non-obesegenic antipsychotic drugs [47]. Also, the high orexin level in patients with schizophrenia treated with antipsychotic drugs is regarded as a protective factor against the development and risk of drug-induced metabolic syndrome [48].…”
