Oral Tolerance Induction inDermatophagoides pteronyssinus-Sensitized Mice Induces Inhibition of IgE Response and Upregulation of TGF-βSecretion

Abstract: Oral antigen administration induces peripheral tolerance in naive animals. Studies of oral tolerance induction in sensitized mice have clinical relevance as a strategy to modulate allergy. In this study, the A/Sn mice sensitized with extract of Dermatophagoides pteronyssinus (Dp) and submitted to oral Dp administration showed a marked decrease in IgE anti-Dp antibody production compared with sensitized phosphate-buffered saline (PBS)-fed mice. T cells from Dp-fed mice cocultured with spleen cells from PBS-fed … Show more

“…Our previous data demonstrated increased TGF-ß basal secretion levels by mesenteric lymph nodes (11), as well as by spleen cells in mice fed with the Dp extract (16). In the present study, a decreased IFN-γ production by peripheral lymph node cells was observed four days after Dp extract feeding.…”

“…to the dust mite Dp extract in mice showed that oral administration down-modulated the Th2-axis, including IgE (11,15), IgG1 and IL-4 production and up-regulated TGF-ß secretion (16). However, the complex regulatory cytokine network triggered by oral tolerance, regarding the specific and natural polyreactive IgA response in the serum and gut secretion of allergen-tolerized mice is not completely understood.…”

“…Oral alloantigen administered continuously just after grafting was more effective than when administered one week after the graft. In another murine model of oral tolerance induction such as type I hypersensitivity to dust mite (14,15), a dose-dependent effect on the modulation of the IgE response was observed in sensitized mice. An additional increase in the allergen dose enhanced the hypersensitivity response, worsening the allergic response (14,15).…”

Similar to the effect obtained with antigen delivery to the mucosal site, injection of donor splenocytes into the portal vein prolonged skin allograft survival.…”

“…TGFβ1 participates in the local regulation of allergen-specific IgE production and peripheral tolerance to allergens [52]. It has been reported that addition of recombinant TGFβ1 to spleen cell cultures stimulated with D. pteronyssinus inhibits IgE secretion [53]. Also oral administration of high-dose TGFβ1 simultaneously with ovalbumin significantly inhibits the elevation of ovalbumin-specific IgE levels and anaphylactic reactions in mice [54].…”
