IntroductionUlcerative colitis (UC) is an inflammatory disease of the intestinal tract with unknown etiology. Both genetic and environmental factors are involved in the occurrence and development of UC. Understanding changes in the microbiome and metabolome of the intestinal tract is crucial for the clinical management and treatment of UC.MethodsHere, we performed metabolomic and metagenomic profiling of fecal samples from healthy control mice (HC group), DSS (Dextran Sulfate Sodium Salt) -induced UC mice (DSS group), and KT2-treated UC mice (KT2 group).Results and DiscussionIn total, 51 metabolites were identified after UC induction, enriched in phenylalanine metabolism, while 27 metabolites were identified after KT2 treatment, enriched in histidine metabolism and bile acid biosynthesis. Fecal microbiome analysis revealed significant differences in nine bacterial species associated with the course of UC, including Bacteroides, Odoribacter, and Burkholderiales, which were correlated with aggravated UC, and Anaerotruncus, Lachnospiraceae, which were correlated with alleviated UC. We also identified a disease-associated network connecting the above bacterial species with UC-associated metabolites, including palmitoyl sphingomyelin, deoxycholic acid, biliverdin, and palmitoleic acid. In conclusion, our results indicated that Anaerotruncus, Lachnospiraceae, and Mucispirillum were protective species against DSS-induced UC in mice. The fecal microbiomes and metabolomes differed significantly among the UC mice and KT2-treated and healthy-control mice, providing potential evidence for the discovery of biomarkers of UC.

Periodontitis is a biofilm‐induced, host‐mediated inflammatory disease that results in periodontal tissue destruction. The design of functional biomaterials based on disease pathophysiology is essential for enhancing their therapeutic effects in periodontitis treatment. As promising localized drug delivery systems and tissue engineering scaffolds, hydrogels have gained significant interest for controlled and sustained release of bioactive agents in periodontal applications. The rational design of bioactive hydrogels can facilitate bacterial control and modulate destructive host inflammation, thereby preventing the progression of periodontitis. In this review, the pathophysiological mechanisms underlying periodontitis as fundamental principles for the design of functional hydrogel systems are first introduced. In the following part, an overview is systematically provided of the types and functions of the bioactive hydrogel systems loaded with anti‐bacterial and anti‐inflammatory agents for periodontal delivery. Finally, the remaining challenges and future perspectives of hydrogel delivery systems for periodontal applications are proposed. It is believed that this review will inspire the rational design and development of innovative functional hydrogel biomaterials toward periodontal therapy.

Intestinal diseases like inflammatory bowel disease (IBD) and colorectal cancer originate from inflammation and disruption of mucosal barriers. Polyphenols can mitigate intestinal inflammation through antioxidant, anti‐inflammatory, and microbiome modulation effects. However, the poor solubility and stability of polyphenols restrict therapeutic delivery. Self‐assembly provides a nanoscale platform to overcome these limitations. Polyphenol‐based nanoparticles (PNPs) are formed via coordination of polyphenols with metals like iron, copper, and zinc based on the catechol/galloyl groups. Templeted assembly with amphiphilic block copolymers can also direct polyphenol self‐assembly into nanostructures. PNPs prepared by these mild, aqueous methods exhibit enhanced stability, pH‐responsive disassembly, high cargo‐loading capacity, and targeted accumulation in inflamed intestinal tissues. PNPs can load with hydrophobic polyphenols, drugs, genes, proteins, or probiotics and demonstrate therapeutic potential in preclinical IBD, colorectal cancer, and microbiome disorder models. Ongoing challenges include augmenting prebiotic effects, multidrug encapsulation, and engineering PNPs as biotherapeutics. Future directions involve tailored polyphenol–polymer covalent assemblies and investigating PNPs interactions with enterocytes, immune cells, and microbiota. Overall, PNPs prepared by facile self‐assembly combine the bioactivities of polyphenols with advanced delivery functionality, presenting new opportunities for combination and microbiota‐based therapies for complex intestinal diseases.
