Polyinosinic:polycytidylic acid [poly (I:C)] was administered in vivo to Chinook salmon (Oncorhynchus tshawytscha) post‐smolts to determine the immune responses on haematological and cellular functional parameters, including spleen (SP), head kidney (HK) and red blood cell (RBC) cytokine expression, as well as serum metabolomics. Poly (I:C) in vivo (24 h exposure) did not affect fish haematological parameters, leucocyte phagocytic activity and phagocytic index, reactive oxygen species and nitric oxide production. Gas chromatography–mass spectrometry–based metabolomics revealed that poly (I:C) significantly altered the serum biochemistry profile of 25 metabolites. Metabolites involved in the branched‐chain amino acid/glutathione and transsulphuration pathways and phospholipid metabolism accumulated in poly (I:C)–treated fish, whereas those involved in the glycolytic and energy metabolism pathways were downregulated. At cytokine transcript level, poly (I:C) induced a significant upregulation of antiviral ifnγ in HK and Mx1 protein in HK, SP and RBCs. This study provides evidence for poly (I:C)–induced, immune‐related biomarkers at metabolic and molecular levels in farmed O. tshawytscha in vivo. These findings provide insights into short‐term effects of poly (I:C) at haematological, innate and adaptive immunity and metabolic levels, setting the stage for future studies.

In previous study, the effects of synthesized chitosan/zeolite and nanochitosan/zeolite composites on rainbow trout growth, digestive enzyme activities and some biochemical parameters were shown. In this study, the effects of experimental diets based on the previous study on the rainbow trout intestinal histology and stereology were assessed: control diet (no zeolite and composite), T1 diet (14.28 g/kg zeolite), T2 diet (0.05 g/kg chitosan included in zeolite), T3 diet (0.5 g/kg chitosan included in zeolite), T4 diet (5 g/kg chitosan included in zeolite), T5 diet (0.05 g/kg nanochitosan included in zeolite), T6 diet (0.5 g/kg nanochitosan included in zeolite), T7 diet (5 g/kg nanochitosan included in zeolite). The experiment was conducted for 60 days. Results showed that supplemental diets did not have side effect on the normal structure of intestinal segments but administration of T1 and T4 diets had slight negative effects on structural maintenance in the middle part of intestine. Acidic mucin producing goblet cell percentage was also higher in nanocomposites‐administrated groups in comparison with the control group. Enhanced villus height, density and finally absorption surface area in different parts of rainbow trout intestine were mainly shown in T7 group. Meanwhile, higher thickness of tunica mascularis in different regions were noted in treatment groups especially in T7 group. The number of intraepithelial mononuclear leukocytes in fish intestine received treatment diets was also higher than control group. In conclusion, nanochitosan/zeolite composites in comparison with zeolite and chitosan/zeolite composites were more effective to improve histological structure of rainbow trout intestine.

Growth hormone-releasing peptide-6 (GHRP-6) is one of the earliest developed synthetic peptidyl growth hormone secretagogue receptor agonists. These compounds mimic the effect of the endogenous ligand ghrelin. In vertebrates, ghrelin is a potent circulating orexigenic hormone with functional roles in controlling food intake, energy expenditure, adiposity, growth hormone secretion and immunity. Ghrelin has been studied mainly in vertebrates; thus, little is known about its role in invertebrates, including crustaceans. We first evaluated the effect of GHRP-6 injection over feed intake in shrimp and its effects on shrimp growth when the peptide was administrated by successive immersion baths. GHRP-6 increased feed intake, body weight and size, the number of rostral spines and gill branches, protein concentration and haemocyte number in treated shrimps. We also evaluated the peptide uptake and clearance in a pharmacokinetics, using [H 3 ]GHRP-6 administered to postlarvae. Given a limited exposure and efficient clearance of the peptide-associated radioactivity from larvae, our findings suggested that GHRP-6-treated Litopenaeus vannamei can be consumed safely by humans after aquaculture applications. These results propose that GHRP-6 could be an additional tool to study growth physiology in crustaceans and also a promising candidate for development into a new biotechnology product for improving shrimp growth and quality.
