Oral Administration of Pregabalin in Rats before or after Nerve Injury Partially Prevents Spontaneous Neuropathic Pain and Long Outlasts the Treatment Period

Abstract: Background and Methods: Pregabalin alleviates stimulus-evoked neuropathic pain (NeuP) in some pain patients and rodents in models of painful neuropathies. But it is not known if pregabalin can also alleviate spontaneous NeuP. Sciatic and saphenous neurectomy in rats elicits spontaneous self-mutilation of the denervated hindpaw, a behavior that models spontaneous NeuP. We tested if pregabalin (20 or 30 mg/kg/day; twice daily, per os) for 7 days before denervation, or 42 days thereafter, can suppress this behavi… Show more

“…Animals were randomly divided into six experimental groups (n 6): control, sham, CCI, CCI treated with the extract (200 and 400 mg/kg) and CCI treated with pregabalin (30 mg/kg) (Wang et al, 2016). Control, sham and CCI groups were given 5 ml/kg (by oral gavage) acacia gum solution (10% w/v), the vehicle used for both the extract and pregabalin.…”

“…Higher preoperative doses and continued use in the postoperative period may provide better analgesia. 3,128 This makes intuitive sense as a 2 d-1 concentrations are raised for several days after injury. 20 Only chronic use was associated with improved ambulation in a knee inflammation model.…”
