Oral Administration of Bovine Lactoferrin-Derived Lactoferricin (Lfcin) B Could Attenuate Enterohemorrhagic Escherichia coli O157:H7 Induced Intestinal Disease through Improving Intestinal Barrier Function and Microbiota

Abstract: Lactoferricin (Lfcin) B, derived from lactoferrin in whey, has attracted considerable attention because of its multiple biological functions. Zoonotic enterohemorrhagic Escherichia coli (EHEC) O157:H7 has adverse effects on intestinal epithelial barrier function, leading to serious intestinal disease. In this study, the EHEC O157:H7-induced intestinal dysfunction model was developed to investigate the effects of Lfcin B on EHEC O157:H7-induced epithelial barrier disruption and microbiota dysbiosis. Results sho… Show more

“…the peptide was prepared in sterile saline and stored under −80 °C until used. Other normal reagents were listed on the paper we have previously published 51 , to introduce briefly: Rabbit polyclonal Abs for zonula occludens (ZO-1), Occludin, CD177 and F4/80 were purchased from Abcam (Cambridge, MA). The secondary Abs used for the immunohistochemistry was goat anti-rabbit IgG conjugated with HRP (Abcam) and for the immunofluorescence were goat anti-rabbit IgG labeling with FITC and Cy3 (Abcam), the TUNEL kit was purchased from Roche (USA).…”

“…Mice (40 in total) with average weight of 20 g were obtained from the Laboratory Animal Center of the Chinses Academy of Sciences (Shanghai, China). Just as the same feeding model we have previously carried out 51 , the mice were individually housed and maintained on a 12:12 h light-dark cycle under specific pathogen free conditions, with free access to feed and water throughout the experimental period. After five days adaptive period, the mice were randomly divided into four groups, each group had ten mice.…”

“…Recently, a therapeutic effect of bovine LFcin after oral intake was reported for the prevention of enterohemorrhagic E. coli (EHEC) infection in a mouse model [59], demonstrating, like numerous other studies, the potential of this antimicrobial peptide in veterinary and human medicine (reviewed in [34]). However, neither different amounts of polySia nor different polySia DPs can influence this ability of LFcin.…”

“…Moreover, the imbalance in intestinal microbiota in functional gastrointestinal disorders (FGIDs) has been proven to increase the level of intestinal endotoxins and the production of pro-in ammatory factors as a result of damage caused to the intestinal mucosal barrier and changes in intestinal permeability, which lead to chronic low-grade in ammation and a decrease in the pH of the intestinal environment [24,25]. In our experiments, no differences in the bacterial phyla were found between HSP patients and healthy children, but signi cant changes in composition and structure were detected between gut microbial families of HSP patients and controls.…”
