Oral administration of Bifidobacterium bifidum TMC3115 to neonatal mice may alleviate IgE-mediated allergic risk in adulthood

Abstract: This study aimed to demonstrate whether exposure to bifidobacteria during early life influences immunity and alleviates the risk of immunoglobulin E (IgE)-mediated allergies in adulthood. BALB/c neonatal mice (n=54) were administered with a lyophilised cell preparation of Bifidobacterium bifidum TMC3115 (TMC3115) for 3 weeks. Following the intervention, the mice were immunised with intraperitoneal ovalbumin (OVA). The morphology and function of the intestinal epithelium were determined using histopathological … Show more

“…The results were consistent with the previous report Bifidobacterium species inhibits IgE production in the immune modulation (Takahashi et al., 2006). Recent work also demonstrated that oral administration of B. bifidum TMC3115 may alleviate IgE‐mediated allergic risk in an animal model (Cheng et al., 2018). On the contrary, IgG2 as IgG subclass may monitor allergen immunological activity (Movérare et al, 2017).…”

“…Comparing with the present work, the effects of B. bifidum TMC3115 on allergy were mostly limited to animal study. Oral administration of B. bifidum TMC3115 in neonatal mice was found to attenuate IgE‐mediated allergic risk in adulthood (RY Cheng et al., 2018). B. bifidum TMC3115 consumption in neonatal mice affected gut microbiota and immunity in adulthood (Cheng et al., 2019).…”

“…Fecal microbiota communities were determined by 16S rRNA sequencing using universal primers (forward primer, 338F, 5′-ACTCCTACGGGAGGCAGCAG-3′; reverse primer, 806R, 5′-GGACTACHVGGGTWTCTAAT-3′) [8]. PCR amplification was performed in a 25 μl reaction mixture containing 50 ng of template DNA, 12.5 μl of Phusion Hot Start Flex 2× Master Mix (New England Biolabs Inc., Beverly, MA, USA), and 2.5 μl of each primer.…”

“…The bioinformatics analysis was performed as described previously [8]. Briefly, the sequencing data were filtered, and the effective tags were clustered into operational taxonomic units (OTUs) with a similarity of 97% using the Uparse 7.0.1001 software (https://drive5.com/uparse/).…”
