Optogenetic dissection of roles of specific cortical interneuron subtypes in GABAergic network synchronization

Abstract: In the presence of the A-type K channel blocker 4-aminopyrdine, spontaneous synchronous network activity develops in the neocortex of mice of either sex. This aberrant synchrony persists in the presence of excitatory amino acid receptor antagonists (EAA blockers) and is considered to arise from synchronous firing of cortical interneurons (INs). Although much attention has been given to the mechanisms underlying this GABAergic synchrony, the contribution of specific IN subtypes to the generation of these long-l… Show more

“…A). In contrast, in the 4AP model, application of glutamate receptor blockers only blocked the ictal‐like activity, but interictal activity persisted, as has been previously reported (Avoli et al, ; Bohannon and Hablitz, ). This interictal activity was eliminated through the application of the of GABA A receptor blocker gabazine (Fig.…”

“…As expected, application of AMPA and NMDA receptor blockers resulted in the abolishment of all interictal and ictal-like activity in the 0 Mg 2+ model (Figure 4A). In contrast, in the 4AP model, application of glutamate receptor blockers only blocked the ictal-like activity, but interictal activity persisted, as has been previously reported 10,31 . This interictal activity was eliminated through the application of the of GABA A receptor blocker gabazine (Figure 4B), strongly suggesting that this remaining epileptiform activity is due to post-synaptic IPSCs onto the pyramidal cells 10 .…”

“…6 In some models, optogenetic activation of interneurons alone is sufficient to initiate seizures. 7 In the highlighted study, Tryba et al 8 use data ranging in scale from cell culture to human electroencephalogram (EEG) to develop a model aimed at describing sometimes the dichotomous role of GABA in an epileptic brain. They form initial hypotheses based largely on simultaneous recordings of neuronal spiking, local field potential (LFP), and EEG from patients with epilepsy.…”
