Optimum Hole-Opening Condition for Cisplatin Incorporation in Single-Wall Carbon Nanohorns and Its Release

Abstract: We enclosed cisplatin (CDDP), an anticancer drug, inside single-wall carbon nanohorns (SWNH) with holes opened by being heated from room temperature to a target temperature (475-580 degrees C) in flowing dry air, with an increase rate of 1 degrees C/min. The optimum target temperature was found to be 500 degrees C, in terms of the least amount of CDDP deposited outside the SWNH, when the quantity of CDDP encapsulated inside the SWNH was 12 wt %. The incorporated CDDP was slowly released from the SWNH in phosph… Show more

“…The exterior and interior pores inherent to SWNHs can be tailored for loading specific drugs by controlling their synthesis process. SWNHs can release controlled amounts of chemotheraputic drugs into tumors to effectively treat cancer [26,27]. The controllable release, ability to store chemotheraputic drugs, and absorption characteristics make SWNHs a strong candidate for both drug delivery and photothermal treatment.…”

“…Intratracheal instillation tests found some incidences of foamy macrophages and anthracosis, but neither resulted in increased mortality rates [21,25]. Furthermore, the extensive surface area, multitude of interstices inherent to SWNHs, and accessible internal volume permit the addition of guest molecules [26]. This makes them an exceptional nanoplatform for drug delivery [18,19].…”

“…6A). If carbon nanohorns are used (carbon nanotubes with rounded ends that cap the particles), then holes can be created in the tubes by heating them up to 500 8C before the drug is loaded (Ajima et al 2006). Release of platins from the nanotubes is controlled either through diffusion or through the use of iron nanoparticle caps A schematic diagram showing a unique method of attaching a platin drug to the surface of gold nanoparticles.…”

“…Here, Cisplatin (Cis-Diamminedichloroplatinum (CDDP)) has been used. The inclusion of Cisplatin has already involved a new variety of SWCNTs: the single-wall carbon nanohorns (SWCNHs), graphitic tubes with cone shaped tips and the characteristic feature to assemble in spherical aggregates [5][6][7].…”
