Objective: The aim of this work was to evaluate the stability and determine the kinetic parameters of degradation of biomarkers kaempferol and quercetin, present in the pre-formulated of the extract of Poincianella pyramidalis obtained by a spray dryer. Methods:A 2 3 experimental design coupled with RSM was applied to evaluate and optimize the effects of processing parameters on the content of chemical markers in dry extracts by a spray dryer. Stability testing was performed to verify the influence of temperature on the degradation of kaempferol and quercetin present in the pre-formulated. The markers contents were determined by HPLC.Results: Surface response analysis showed the influence of the independent variables on the responses of the concentration kaempferol and quercentin biomarkers on the process. The variables of the inlet air temperature, flow feed rate and the adjuvant ratio presented negative responses with significant difference (p<0.05). According to the data obtained in the stability of the pre-formulated studied zero and second orders kinetics models the for degradation of the kaempferol and only second order kinetic model for the quercetin. It was also evaluated reducing the concentration of both biomarkers studied throughout the study. Conclusion:In the present study, it was observed that all independent variables of the drying process by spray dryer showed the greatest influence on the concentration of the studied markers. Two markers had a different thermal behavior compared to the different excipients studied and there was degradation of both the quercentin biomarker and kaempferol during the study period.

Vitamin A palmitate (VAP) contains retinol and palmitic acid which is easily absorbed by body and widely used in skin care products. But, it is a hydrophobic and oxidation sensitive molecule which undergoes rapid degradation especially in an aqueous environment. The purpose of this study was to prepare microcapsules of VAP using combination maltodextrin and modified starches. Emulsion of VAP was prepared using cremophore RH 40 with Tween 80 in a homogenizer and formed emulsion was spray-dried. The spray process was optimized using a central composite design for two variables to obtain microcapsules with desirable characteristics. Microcapsules containing 30% of VAP were produced using different concentration of wall materials. The prepared microcapsules were evaluated for their physical, morphological, in-vitro drug release and SEM study. The results showed that obtained microcapsules are nearly spherical in shape with a particle size ranged from 1 to 12 μm. The drug content and encapsulation efficiency (53% -63%) of different batches were found within acceptable range. These stabilized drug loaded microcapsules were incorporated into silicone cream based formulation for convenient topical application and evaluated for its physicochemical parameters. The drug release study showed 80.18% to 83.43% of drug release from VAP microcapsules while topical formulations prepared by VAP microcapsules showed 67.09% to 71.45% drug release at the end of 24 hrs. The formulations were kept for 3 months stability study as per ICH guidelines and found to be stable.

To explore the potential use of Guava as a functional food ingredient, aqueous guava extract was filtered to remove insoluble material; the soluble extract was spray-dried with the addition of 7% maltodextrin. Spray dried Guava concentrate showed increased levels of antioxidant capacity (2, 2-diphenyl-1-picrylhydrazyl DPPH assay 3.2 fold) and total phenolics (2.0 fold) but low levels of flavinoids. The addition of maltodextrin during spray-drying resulted in the formation of certain unresolved peaks analyzed by HPLC due to dilution effect, the redistribution of molecular weight is likely beneficial for in vivo absorption and increase its bioavailability. Spray-drying concentration also increased the proportion of ascorbic acid content (118.6 mg/100g) of guava powder and showed effective antimicrobial activity against Shigella (MIC 11mg/ml), E coli (MIC 8 mg/ml) and Candida spp. (MIC<1 mg/ml). The proximate and physiochemical properties showed good quality and increased solubility of guava powder compared to plain guava extract. Overall the spray-dried Guava powder may be a good source of natural antioxidants and profoundly increase use of Guava in value added processing and dietary intake.
