Optimization of the reconstruction of dermal papilla like tissues employing umbilical cord mesenchymal stem cells

Abstract: Alopecia is not life threatening, but patients who undergo alopecia often experience severe mental stress. In addition, the number of individuals afflicted by alopecia has been increasing steadily. The most effective treatment of alopecia developed to date is auto hair transplantation. To overcome the limitations associated with current therapies for the treatment of alopecia, many researchers have attempted to revive hair follicles by áå=îáíêç culture of hair follicle cells and subsequent implantation in the … Show more

“…The Korean team of Yoo et al [ 87 89 ] examined the effect of hWJSC on the acceleration of wound healing processes along with formation of hair follicles and other skin appendages. Enriched aggregates of hWJSC cells can form new hair follicles.…”

“…The addition of growth factors to the culture medium is required: hepatocyte growth factor (HGF) stimulates growth of hair follicles in vivo and in vitro ; basic fibroblast growth factor (bFGF) stimulates growth of dermal papilla cells in vivo ; and vesicular endothelial growth factor (VEGF) stimulates growth of hair follicles and hair root in vivo . The hepatocyte growth factor (HGF) must be used at the stage of differentiation of the dermal papilla in culture, which is relatively expensive [ 87 89 ].…”

“…Out of these, auto hair transplantation has been the most effective. In addition, because of critical limitations due to cost, amount of hair and number of operations (4), studies on hair regeneration have been attempted using cultured hair follicle cells or mesenchymal stem cells (4)(5)(6)(7)(8).…”

“…However, DP isolation is technically laborious and difficult, and cultured dermal papilla cells (DPCs) lose their hair inductive abilities after a few passages. To overcome these limitations, studies on the reconstruction of dermal papilla-like tissues have been performed with human umbilical cord-derived mesenchymal stem cells (4,5,7).…”

“…These cells may be an attractive therapeutic tool for regenerative medicine and tissue engineering (Aggarwal and Pittenger, 2005; Risbud and Shapiro, 2005; Yoo et al, 2010). Therefore, more recent studies have focused on adult stem cells for future clinical applications (Crop et al., 2009; Gaebel et al, 2011).…”
