Optimization of an Extraction Solvent for Angiotensin-Converting Enzyme Inhibitors from Hibiscus sabdariffa L. Based on Its UPLC-MS/MS Metabolic Profiling

Abstract: Hibiscus species (Malvaceae) have been long used as an antihypertensive folk remedy. The aim of our study was to specify the optimum solvent for extraction of the angiotensin-converting enzyme inhibiting (ACEI) constituents from Hibiscus sabdariffa L. The 80% methanol extract (H2) showed the highest ACEI activity, which exceeds that of the standard captopril (IC50 0.01255 ± 0.00343 and 0.210 ± 0.005 µg/mL, respectively). Additionally, in a comprehensive metabolomics approach, an ultra-performance liquid chroma… Show more

“…By contrast, ferulic acid and its derivatives were less reported as constituents of Hibiscus spp. extracts, but they may be of great importance for their biological activities [ 6 , 42 , 43 , 44 ]. Indeed, ferulic acid derivatives obtained from different Hibiscus species showed important pharmacological properties such as antiviral and angiotensin-converting enzyme inhibitory activities [ 43 , 44 ].…”

Indeed, ferulic acid derivatives obtained from different Hibiscus species showed important pharmacological properties such as antiviral and angiotensin-converting enzyme inhibitory activities [ 43 , 44 ]. In addition, ferulic acid was described as an active molecule in H. mutabilis , H. taiwanensis extracts [ 45 , 46 ], and in H. sabdariffa calyx extracts [ 28 , 38 ].…”

“…The mass spectra were acquired in the positive and negative ionization mode using a heated electrospray ionization (HESI) source in combination with an Orbitrap-type HRMS ( Salem et al, 2016 ). Metabolites were identified by their mass spectra and comparison with our in-house database and the reference literature reports ( Salem et al, 2020 ). All the obtained data were analyzed and correlated using MetaboAnalyst ( Pang et al, 2020 ).…”

“…Figuring out the mystery of whether the hot or cold extract possesses antihypertensive potential is essential. Thus, we present herein a comparative metabolomics analysis of cold and hot aqueous extracts of Hibiscus sabdariffa L. with respect to their antihypertensive potentials in light of continuing our work on the best extraction method for Hibiscus antihypertensive metabolites ( Salem et al, 2020 ). To accomplish such a goal, an in vitro assay was initially performed to evaluate the inhibitory activity of Hibiscus metabolites on ACE and renin activities.…”

“…Roselle (H. sabdariffa L.) is a plant from the Malvaceae family and is widely grown in Indonesia. People use roselle calyx as herbal tea because it contains several substances such as phenolic acids, organic acids, fatty acids, flavonoids and anthocyanins (Salem et al, 2020) that giving roselle antioxidant properties. Several studies have shown that roselle effectively lowers blood pressure, lowers fat profile, has antiinflammatory, antimicrobial, anti-anaemia, anticarcinogenic, hepatoprotective, and improves fertility disorder (Riaz and Chopra, 2018).…”
