Optimisation of a gold nanoparticle-based aptasensor integrated with image processing for the colorimetric detection of acephate using response surface methodology

Abstract: We successfully optimized AuNPs, modified DNA aptamer and magnesium sulphate salt to enhance the selectivity and sensitivity for detection of Ac. The accuracy of the detection was also improved by image processing technique.

“…In order to detect acephate, Osman MJ et al proposed an image processing technology combined with an aptamer sensor, using the sensor to obtain the relevant information of the aptamer, and using the image processing technology to achieve colorimetric detection, and finally proved in the experiment the effectiveness of the current method. Although this research can effectively achieve image detection, it uses sensor technology, which is vulnerable to environmental interference [13]. In order to reduce the mite infection in bee farming, Sevin S et al proposed an image capture device that can automatically focus detection.…”

“…Qualities, like, simple operations, quick response, and easy pre-treatment of samples for analysis make biosensors relevant for monitoring the acephate residues. Several biosensors employing different transducing mechanisms, such as chemiluminescence [8], uorescence [9], colorimetry [10], and electrochemical [11] have been developed for the detection of acephate. Electrochemical techniques, such as differential pulse voltammetry (DPV), chronoamperometry (CA) [12], and electrochemical impedance spectroscopy (EIS) [13], have previously been utilized for the detection of acephate.…”

“…The recognition elements employed in these biosensors have broad speci city therefore they can identify organophosphate pesticides rather than being selective to acephate only. The colorimetric biosensor developed by Junaedy Osman et al [10] utilizes a broad speci c aptamer, which again may produce response in presence of other pesticides. Therefore, using an aptamer speci cally selected for acephate recognition enhances the ability to selectively sense acephate.…”

“…Prior attempts to optimize NP synthesis have been reported utilizing different approaches with varying optimization goals, but most primarily focus on a single response: either optimizing the yield or the activity. 15–18 To the best of the authors' knowledge at the time of writing this article, simultaneous optimization of both synthesis yield and activity of produced NPs is currently not present in the literature. For example, a recent, comprehensive review by Rodrigues et al concerning the optimization of noble metal nanoparticles as nanocatalysts did not reveal any prior dual-optimization approaches.…”
