Optimal Stem Cell Transporting Conditions to Maintain Cell Viability and Characteristics

Abstract: BACKGROUND: The preservation of stem cell viability and characteristics during cell transport from the bench to patients can significantly affect the success of cell therapy. Factors such as suspending medium, time, temperature, cell density, and container type could be considered for transport conditions. METHODS: To establish optimal conditions, human amniotic fluid stem cells' (AFSCs) viabilities were analyzed under different media {DMEM(H), DMEM/F-12, K-SFM, RPMI 1640, a-MEM, DMEM(L), PBS or saline}, tempe… Show more

“…Compared with the PLGA group, the values for both cell groups were significantly lower. Thus, the functional analysis confirmed that the Cells transported under conditions established in an in vitro experiment [3] showed recovery of total kidney volume, regeneration of glomerular/renal tubules, and low CD4/CD8 positive T cell infiltration. In addition, migration to distant organs and tumorigenicity were not observed over 40 weeks.…”

“…To minimize these problems, in a previous study we optimized the cell transport conditions [3] through an in vitro analysis of the medium, temperature, cell density, and container type. For AFSCs, we suggested an appropriate cell suspension medium comprising high-glucose Dulbecco's modified minimal Eagle's medium (DMEM), a transport temperature of 4 °C, a cell density of 1 9 10 7 / 2 mL, and a disposable syringe as the container.…”

“…It was shown that IGF-2 promoted the adipogenic differentiation of stem cells at 100 ng/mL concentration [26]. Likewise, the net effects or optimal dosage may differ between the studies due to culturing conditions, type, and passages of cells [27]. Moreover, IGF-2 may have additional functions.…”

“…8 The cell-based therapies require the transportation of cells to the targeted locations for transplantation. 9 This transportation and delivery need to be precise, and a failed cell delivery could cause a serious immune response. 10 Microrobots are very suitable candidates to perform this crucial step for delivering cells accurately and noninvasively.…”
