Optical Brain Imaging: A Powerful Tool for Neuroscience

Abstract: As the control center of organisms, the brain remains little understood due to its complexity. Taking advantage of imaging methods, scientists have found an accessible approach to unraveling the mystery of neuroscience. Among these methods, optical imaging techniques are widely used due to their high molecular specificity and single-molecule sensitivity. Here, we overview several optical imaging techniques in neuroscience of recent years, including brain clearing, the micro-optical sectioning tomography system… Show more

“…The US-retrieval combined with β-AR antagonism could be a potential strategy for the treatment of memory disorders, such as substance addiction or post-traumatic stress disorders. The neuronal circuitry underlying memory reconsolidation triggered by US-retrieval or CS-retrieval should be studied in our future research (Zhu et al, 2017 ).…”

“…Meanwhile, over 10 different clearing agents are in use (for review, see Refs. 9 , 11 , and 34 ). While a suitable and effective clearing method was essential for LSFM in the case of nonexpanded samples to achieve the required transparency, this aspect became irrelevant when combining it with expansion to LSFEM, because expanded samples were intrinsically transparent due to their high water content.…”

“…The diffusion barrier properties caused by the lipid bilayer of cells are related to chemical penetration and light scattering properties of the lipid-water interface, making the tissue difficult to visualize [5,24,32]. Therefore, lipid extraction is critical for the optical transparency of tissue.…”

“…Biological tissue is a three-dimensional structure with complex detailed structures and vast molecular information, and how to acquire these data has always been a critical challenge [1][2][3][4][5][6][7][8]. This task has mainly relied on cutting the tissue into thin slices and provide cellular information at the two-dimensional level.…”
