Opposite extremes in ethylene/nitric oxide ratio induce cell death in suspension culture and root apices of tomato exposed to salt stress

Abstract: The plant hormone ethylene or the gaseous signalling molecule nitric oxide (NO) may enhance salt stress tolerance by maintaining ion homeostasis, first of all K+/Na+ ratio of tissues. Ethylene and NO accumulation increased in the root apices and suspension culture cells of tomato at sublethal salt stress caused by 100 mM NaCl, however, the induction phase of programmed cell death (PCD) was different at lethal salt concentration. The production of ethylene by root apices and the accumulation of NO in the cells … Show more

“…Disintegration of cell membranes was determined by the measurement of electrolyte leakage [18]. Tissue segments of the same size were collected on a filter paper and washed twice with 50 ml of double distilled water and then 0.5 g of fresh tissues was incubated in 20 ml of double distilled water at 25 °C for 2 hours.…”

“…and H 2 O 2 ) and nitrogen (NO and ONOO -) forms in the root apices of wild type and ethylene receptor mutants and to reveal correlations between their accumulation and cell viability. We are also interested in how various forms of reactive oxygen and nitrogen affect the acclimation to salt stress or the initiation of PCD in Nr mutants exposed to salinity stress which had been found to be sublethal and lethal stress in wild type tomato roots (Poór et al 2014).…”

“…Flow rates were 35 mL min -1 for He, 30 mL min -1 for H 2 and 300 mL min -1 for air. The oven, injector and detector temperatures were 300, 120 and 100 o C, respectively (Poór et al, 2014). A set of ethylene standards was used to calculate the amount of ethylene generated by the root segments.…”
