Opioids for hedonic experience and dopamine to get ready for it

Abstract: Altogether, the data presented point out for an implication of dopamine in the anticipatory/preparatory aspects of feeding more than on the motivational and consummatory aspects. However, dopamine involvement in the anticipatory/preparatory component of feeding seems specifically related to very relevant stimuli, such as highly palatable foods. On the other hand, our data, as well as those present in the literature, strongly suggest a role for opioids in food intake through their modulation of the hedonic perc… Show more

“…Suggestions have included motivational incentive salience, arousal, motivation, and memory consolidation (Barbano and Cador 2007;Berridge 2007;Niv et al 2007;Redgrave and Gurney 2006;Robbins and Everitt 2007;Salamone et al 2007). If so, long-term plasticity in limbic structures, such as synaptic LTP and LTD, might reflect enduring motivation changes amongst other things rather than represent new learned associations via Hebbian synaptic coupling or prediction computation.…”

“…This functional brain network consists of structures such as the amygdala (Gilbert et al, 2003), the insular cortex (Desgranges et al, 2009;Roman et al, 2009) or the parabrachial nucleus (Reilly, 1999;Reilly and Trifunovic, 2000), which are involved in the establishment of a feeding preference or aversion, depending on the sensorial stimuli involved. Literature data also report 'hedonic hotspots' distributed in different brain structures such as the nucleus accumbens (Baldo and Kelley, 2007;Barbano and Cador, 2007;Pritchett et al, 2010), the ventral pallidum (Berridge, 2009) or the subthalamic nucleus (Baunez et al, 2002). The ventral striatum (i.e.…”

“…It is well acknowledged today that the dopamine and opioid systems play an important role in the modulation of feeding behaviour, although they are involved in different steps of this process. These two systems, which are important for the 'reward circuit' and play a major role in food pleasure and selection, have been relatively well characterized in humans and rats (Berridge, 2000;Kelley et al, 2002;Barbano and Cador, 2007;Barbano et al, 2009;Wassum et al, 2009) but not yet in pigs.…”

“…Enkephalins belong to the family of endogenous opioids and are involved in, e.g., reward/ motivation (Barbano and Cador 2007) and the regulation of the stress response (Drolet et al 2001). An involvement of the endogenous opioidergic system in the mediation of several symptoms of depression has long been discussed, and it has been shown that enkephalin catabolism inhibitors (RB101) induce antidepressant-like effects (Roques 2000).…”
