Eight bacterial strains were isolated on kraft lignin (KL) containing mineral salt medium (L-MSM) agar with glucose and peptone from the sludge of pulp and paper mill. Out of these, ITRC-S8 was selected for KL degradation, because of its fast growth at highest tested KL concentration and use of various lignin-related low molecular weight aromatic compounds (LMWACs) as sole source of carbon and energy. The bacterium was identified by biochemical tests as Gram-positive, rod-shaped and non-motile. Subsequent 16S rRNA gene sequencing showed 95% base sequence homology and it was identified as Bacillus sp. In batch experiments, a decrease in pH was observed initially followed by an increase till it reached an alkaline pH, which did not alter the culture growth significantly. The bacterium reduced the colour and KL content of 500 mg l(-1 )KL in MSM, in the presence of glucose and peptone, at pH 7.6, temperature 30 degrees C, agitation of 120 rpm and 6 days of incubation by 65 and 37% respectively. Significant reduction in colour and KL content in subsequent incubations is indicative of a co-metabolism mechanism, possibly due to initial utilization of added co-substrates for energy followed by utilization of KL as a co-metabolic. The degradation of KL by bacterium was confirmed by GC-MS analysis indicating formation of several LMWACs such as t-cinnamic acid, 3, 4, 5-trimethoxy benzaldehyde and ferulic acid as degradation products, which were not present in the control (uninoculated) sample. This favours the idea of biochemical modification of the KL polymer to a single monomer unit.

The pulp and paper (P&P) industry worldwide has achieved substantial progress in treating both process water and wastewater, thus limiting the discharge of pollutants to receiving waters. This review covers a variety of wastewater treatment methods, which provide P&P companies with costeffective ways to limit the release of biological or chemical oxygen demand, toxicity, solids, color, and other indicators of pollutant load. Conventional wastewater treatment systems, often comprising primary clarification followed by activated sludge processes, have been widely implemented in the P&P industry. Higher levels of pollutant removal can be achieved by supplementary treatments, which can include anaerobic biological stages, advanced oxidation processes, bioreactors, and membrane filtration technologies. Improvements in the performance of wastewater treatment operations often can be achieved by effective measurement technologies and by strategic addition of agents including coagulants, flocculants, filter aids, and optimized fungal or bacterial cultures. In addition, P&P mills can implement upstream process changes, including dissolved-air-flotation (DAF) systems, filtration save-alls, and kidney-like operations to purify process waters, thus reducing the load of pollutants and the volume of effluent being discharged to end-of-pipe wastewater treatment plants. A wastewater treatment plant controls the level of pollution reaching the environment. Yet, despite being the most common aerobic route for treatment of wastewater, the activated sludge process is not utilized to its full potential. This is mainly due to the lack of knowledge base correlating the microbial community in the activated sludge to its degradative performance. In this study, the activated biomass at the treatment site was monitored for five consecutive months. Even though operational parameters were kept constant, the microbial community was observed to change after 3 months. This shift was seen to correlate with 25 % loss of degradative efficiency. Target oxygenases were monitored at two time points, and results indicated that the dominating pathway operating in the common effluent treatment plant (CETP) is the degradation of chlorinated aromatics. This study demonstrates the change in degradative efficiency in a CETP with the change in microbial community and analyzes the parameters influencing the microbial community of activated sludge.
