Operation of the Xanthophyll Cycle and Degradation of D1 Protein in the Inducible CAM plant, Talinum triangulare, under Water Deficit

Abstract: Changes in photochemical activity induced by water deficit were investigated in Talinum triangulare, an inducible CAM plant. The aim was to analyse the interactions between C3 photosynthesis, induction and activity of CAM, photosynthetic energy regulation and the mechanisms responsible for photoprotection and photoinhibition under water stress. Gas exchange, chlorophyll a fluorescence, titratable acidity, carotenoid composition and relative contents of the PSII reaction centre protein (D1) were measured. A dec… Show more

“…The supernatant was collected and the pellet extracted again with 100% acetone until it was colourless. Extracts were combined and filtered through a nylon membrane (0.45 mm pore size) and the carotenoids analysed by HPLC as described by Pieters et al (2003). Conversion of peak areas from leaf extracts to pigment amounts was done using pure standards prepared by separation of bands of individual pigments by thin layer chromatography.…”

“…Araure 4, resulting in a 40% increase in DEPS (Pieters and El Souki, 2005). The DEPS state was also shown to increase under drought conditions in Talinum triangulare (Pieters et al, 2003). In recent years, a gene family representing 10-13 independent amino acid sequences belonging to the fibrillin family has been identified in Arabidopsis, tomato and rice (Laizet et al, 2004).…”

“…(Portulacaceae) is a succulent and erect plant, with sessile lanceolate leaves, and a C 3 /crassulacean acid metabolism (CAM) photosynthetic pathway (Guralnick and Jackson , Pieters et al. ). Although cultivated as a leafy vegetable in many parts of Africa, South America and Asia, it is popularly used as medicine to contain many ailments including cancer, diabetes, hepatic disorders, leishmaniasis and reproductive disorders (Jung et al.…”

“…While induced CAM appears to be a common adaptive strategy for many members of the Portulacaceae under different abiotic stress conditions (Guralnick and Jackson , Pieters et al. ), others such as the purslane ( Portulaca oleracea L.) have been shown to adapt to salt stress by an enhanced antioxidative system and proline accumulation (Yazici et al. ).…”
