Onset and dose relationships of ACTH effects on blood pressure in sheep.

Abstract: SUMMARY The threshold and dose-response relationships for the blood pressure and metabolic effects of adrenocorticotropic hormone (corticotropin, ACTH) were examined in conscious sheep. Corticotropin was infused at five rates (0.5 jig/kg/day, n -4; 1 /ig/kg/day, n = 4; 2 /xg/kg/day, n = 6; 5 jxg/kg/day, n = 5; and 10 ^tg/kg/day, n = 5) for 3 days, and the time of onset of the rise in blood pressure was assessed with a computer-based system. The effects of equimolar infusion of /3-endorphin and ACTH at 5 ^ig/kg… Show more

“…Animals were offered 800 g lucerne-oaten chaff daily ([Na] 60-100 mmol kg-') and water ad libitum. On the day of experiment an 18 gauge needle was inserted into the exteriorised left carotid artery for direct measurement and on-line monitoring of blood pressure via a transducer (Scoggins et al, 1985). Glomerular filtration rate (GFR), effective renal plasma flow (ERPF) and proximal tubular function were measured by established clearance procedures (Whitworth et al, 1978;Yates et al, 1988).…”

“…Experiments were carried out in four adult oophorectomized Merino ewes (35-45 kg). Haemodynamic parameters were measured by previously described methods (Scoggins et al 1985) which enabled beat-to-beat computer monitoring of cardiac output, heart rate and mean arterial blood pressure (MAP). Stroke volume and calculated total peripheral resistance (CTPR) were derived from measured variables.…”

“…Following cessation o f ouabain infusion, 0.15 mol 11 saline at 12 ml h'1 was infused for a further 120 min. Blood pressure was monitored throughout the experiment via a 18-gauge needle inserted into the caro tid artery skin loop [14]. GFR was calculated as the clearance oC'Cr-EDTA [13,15].…”
