One‐Step Rapid Fabrication of Cell‐Only Living Fibers

Abstract: Cellular aggregates are used as relevant regenerative building blocks, tissue models, and cell delivery platforms. Biomaterial‐free structures are often assembled either as 2D cell sheets or spherical microaggregates, both incompatible with free‐form deposition, and dependent on challenging processes for macroscale 3D upscaling. The continuous and elongated nature of fiber‐shaped materials enables their deposition in unrestricted multiple directions. Cellular fiber fabrication has often required exogenously pr… Show more

“…Preparation of SH: Polystyrene petri dishes were treated according to previous reports. [39] Briefly, a layer of an air-cured coating agent (WX2100, Cytonix) was created onto petri dishes surfaces. After drying for 48 h, the excess of the sprayed solution was removed and submerged in a solution of 70% ethanol for at least 3 days.…”

“…Newer extrusion techniques with lower speed can produce high-density constructs with radial and axial patterning [ 34 , 35 ] but have difficulty in creating branching networks and require specialized equipment. Alternatively, the hanging drop method has been modified with patterned substrates in rectangularly designed hydrophilic regions to confine cells in a semi-cylindrical fashion to assemble ECM-free fibers [ 36 ]. This interesting approach is an advancement over the traditional hanging drop method but is still limited in its ability to form multicellular patterns radially or axially.…”

“…In an intriguing study, the Mano group recently showed that mammalian cells are also able to form desired three-dimensional structures such as fibers without the need for genetic or chemical modification but simply by providing a sophisticated cell culture environment [ 196 ]. In their work, they placed a hydrophilic, wettable line surrounded by superhydrophobic areas in a culture dish.…”

“…Driven by gravity, the cells accumulated at the bottom and started building intercellular connections. The length and thickness of these fibers could be tuned by the initial cell concentration and it was possible to grow fibers made from several different cell types together [ 196 ]. The Joshi group recently used a similar approach to synthesize stiff living materials [ 198 ].…”
