One‐Pot Preparation of Polymer–Enzyme–Metallic Nanoparticle Composite Films for High‐Performance Biosensing of Glucose and Galactose

Abstract: New polymer–enzyme–metallic nanoparticle composite films with a high‐load and a high‐activity of immobilized enzymes and obvious electrocatalysis/nano‐enhancement effects for biosensing of glucose and galactose are designed and prepared by a one‐pot chemical pre‐synthesis/electropolymerization (CPSE) protocol. Dopamine (DA) as a reductant and a monomer, glucose oxidase (GOx) or galactose oxidase (GaOx) as the enzyme, and HAuCl4 or H2PtCl6 as an oxidant to trigger DA polymerization and the source of metallic na… Show more

“…16 In addition to its fascinating adhesive properties, dopamine also possesses a remarkable reducing character, which can be used as an eco-friendly and effective reducing agent to fabricate polymer nanocomposites. 19 In Asia, the sacred lotus (Nelumbo nucifera) has been a symbol of purity for over 2000 years. Water droplets can roll freely in all directions and then pick up dirt particles on lotus leaves, resulting in the so-called self-cleaning effect or lotus effect (Fig.…”

“…1C, only a small number of Au NPs were observed on the PDA-coated PDMS surface. As is well known, DA is a reductant and a monomer, and HAuCl 4 or HPtCl 6 can be utilized as an oxidizing reagent to trigger DA polymerization and the source of metallic nanoparticles [29]. After filling the mixture of DA and HAuCl 4 (molar concentration ratio of 7.5:1) into the PDMS microchannel and reacted for 30 min, it is clear that many densely dispersed nanoparticles with the size of $40 nm are homogeneously produced inside and outside the PDA film without aggregation (Fig.…”

“…Then, a freshly prepared 0.1 M phosphate solution containing DA and HAuCl 4 (molar concentration ratio of 7.5:1) at pH 7.17 was filled into the microchannel and reacted for 30 min [29]. After the formation of PDA/Au NP coating, the solution in the channel was flushed out and the PDA/Au NP-modified microchannel was rinsed with water for 5 min to remove physical attachment of residuary DA on the channel surface.…”

“…[39] In enzymes catalysis field, dopamine can be utilized to fabricate new polymer-enzyme-metallic nanoparticle composite films with a high load and a high-activity of immobilized enzymes for biosensing of glucose and galactose. [40] Simultaneously, the dopamine can coat magnetic nanoparticles onto cell surfaces during the self-polymerization for facilitating the use of cells in biocatalysis ( Figures 6A and 6B). [41] Based on the same principle, a polydopamine intermediate was used in the fabrication of 3-layer organic film on silicon wafer by a multistep self-assembly process.…”
