Oncostatin M receptor, positively regulated by SP1, promotes gastric cancer growth and metastasis upon treatment with Oncostatin M

Abstract: Background Oncostatin M receptor (OSMR) is a member of the interleukin 6 (IL-6) receptor family that transduces signaling events of Oncostatin M (OSM). OSM-OSMR signaling plays a key role in inflammation and cancer progression. However, the role of OSM-OSMR in gastric cancer (GC) is still unknown. Methods OSMR expression in GC was determined by real-time PCR (RT-PCR), immunohistochemistry (IHC) and Western blot. The effects of OSM-OSMR on GC cell proliferation, migration, invasion, and epithelial-mesenchymal t… Show more

“…OSM, another IL-6 cytokine family member, is crucial for JAK/STAT activation. It interacts with the extracellular matrix (ECM) proteins, and OSM/OSM receptor (OSMR)-mediated JAK2/STAT3/FAK/Src activation promotes EMT and CSC production [34][35][36][37][38][39]. Other pro-inflammatory cytokines, IL-32β and IL-23, are highly expressed in cancers.…”

“…IHC staining was performed on the tissue microarray according to a previously reported standard protocol used in Shanghai Institute of Digestive Surgery [36]. In brief, freshly cut 5-μm-thick tissue microarray was stained with antibodies against CTSL (1:200, ab203028, Abcam, Cambridge, MA, USA), VEGF-D (1:1000; 26915-1-AP; ProteinTech Group, Chicago, IL, USA) and PECAM1 (1:200, ab268100; Abcam, Cambridge, MA, USA).…”

“…The crystal structure of OSM reveals that the protein comprises four α helices ranging from 15 to 22 amino acids in length (termed A, B, C, and D) and linked by polypeptide loops (Figure 1A) (Deller et al, 2000). The OSMR is a member of the IL-6 receptor family that transduces signaling events of OSM (Yu et al, 2019). Currently, available antibodies, such as GSK315234 and GSK2330811, have already been proven to affect the OSM signal (Verstockt et al, 2019).…”
